Some polyphenol-derived metabolites reach human breast cancer (BC) tissues at concentrations that induce cell senescence. However, this is unknown for isoflavones, curcuminoids, and lignans. Here, their metabolic profiling in normal (NT) malignant (MT) mammary of newly-diagnosed BC patients the tissue-occurring metabolites' anticancer activity are evaluated.Patients (n = 26) consumed 3 capsules/day (turmeric, red clover, flaxseed extracts plus resveratrol; 296.4 mg phenolics/capsule) from biopsy-confirmed diagnosis to surgery (5 ± 2 days) or did not consume capsules 13). NT MT, blood, urine analyzed by UPLC-QTOF-MS using targeted metabolomics. Anticancer was tested MCF-7 MDA-MB-231 cells. Mainly phase-II were detected (108, 84, 49, 47 urine, plasma, NT, respectively). Total metabolite reached 10.7 11.1 2.5 2.4 µmol L-1 respectively. Free curcumin, but its glucuronide, (1.1 1.8 0.2 Breast antiproliferation mainly exerted p53-wild-type cells curcuminoids through cycle arrest, senescence, apoptosis induction via p53/p21 induction, while isoflavone-derived estrogenic-like activity.Curcuminoids could be coadjuvants might help fight upon regular consumption.

Ellagitannins (ETs) and ellagic acid (EA) are dietary polyphenols poorly absorbed but extensively metabolized by the human gut microbiota to produce different urolithins (Uros). Depending on individuals' microbial signatures, ETs metabolism can yield Uro metabotypes A, B, or 0, potentially impacting health after consuming ETs. Human evidence points improved brain ET-rich foods, mainly pomegranate juices extracts containing punicalagin, punicalin, EA-derivatives. Although (or) EA necessary exert effects, precise mechanism, actual metabolites, final drivers responsible for observed effects have not been unraveled.
