Women with obesity usually need larger doses of FSH for ovarian stimulation, resulting in poor outcomes; however, the mechanism is still unclear. To investigate molecular regulation receptor (FSHR) expression associated obesity. Case-control study to improve vitro fertilization (IVF) outcomes. (82) and women who were overweight (457) undergoing IVF 1790 age-matched controls normal weight from our reproductive medicine center. FSHR was decreased parallel body mass index (BMI), whereas estradiol (E2) level on human chorionic gonadotropin (hCG) trigger day significantly lower. granulosa cells (hGCs), both mRNA (P = 0.02) protein 0.001) levels, or obese. Both insulin < glucose 0.0017) levels positively correlated BMI fasting blood follicle fluids (FFs) but not FFs leptin level. We treated granulosa-like tumor (KGN) insulin; E2 production compromised; phosphorylated (p)-protein kinase B (p-Akt2) decreased, p-glycogen synthase 3 (GSK3) increased; there similar changes hGCs Stimulated compound 21 (CP21), an inhibitor GSK3β, resulted upregulated β-catenin activation increased expression. CP21 also substrate 1 phosphatidylinositol 3-kinase (PI3K), as well p-Akt2. reduced caused by a dysfunctional pathway. Decreased insulin-treated KGN could be rescued which might potential target improvement impaired FSH-stimulation response

Ascidians are the closest living relatives of vertebrates, and their study is important for understanding evolutionary processes oocyte maturation ovulation. In this study, we first examined ovulation Ciona intestinalis Type A by monitoring follicle rupture in vitro, identifying a novel mechanism neuropeptidergic regulation vasopressin family peptide (CiVP) directly upregulated phosphorylation extracellular signal–regulated kinase (CiErk1/2) via its receptor. CiVP ultimately activated maturation-promoting factor, leading to germinal vesicle breakdown. CiErk1/2 also induced expression matrix metalloproteinase (CiMMP2/9/13) oocyte, resulting collagen degradation outer follicular cell layer liberation fertile oocytes from ovary. This demonstration essential pathways regulating ascidians will facilitate investigations process peptidergic throughout phylum Chordata.

Rationale: Zearalenone (ZEN), a pollutant in our daily diet, seriously threatens the reproductive health of humans and animals. The primordial follicle (PF) assembly mouse occurs during perinatal period, which determines whole ovarian reserve lifespan. In current investigation, we administered ZEN orally to mice from 16.5 days post coitum (dpc) postnatal day 3 (PD3), single-cell RNA sequencing (scRNA-seq) was performed on PD0 PD3 tissues offspring check toxic formation at single cell level. Methods: Ovarian (in vivo) were examined by analysis, Immunostaining, Western blotting. vitro) qRT-PCR, Results: We found that exposure altered developmental trajectory both germ cells granulosa cells. Furthermore, after establishing cell-cell communication network between cells, this perturbed exposure, especially Hippo signaling pathway. Conclusions: This study showed affected status through pathway blocked PFs. research contributes deeper understanding mechanisms toxicity different types disruption normal intercellular exposure.
