Abstract Expanded polytetrafluoroethylene (ePTFE) is commonly used for vascular grafts. However, its application as small‐diameter grafts has been hampered due to problems such thrombosis and restenosis stemming from mismatched mechanical properties lack of endothelialization. As an effort fabricate with akin natural blood vessels, extra expansion step in the fabrication ePTFE was introduced that led a more compliant double‐expanded PTFE (dePTFE). Further, layer P(AAm‐co‐NaAMPS)‐xanthan‐alginate hydrogel synthesized mechanically interlocked onto luminal surface dePTFE In this novel synergistic approach, outer polymer provided support while inner enhanced cytocompatibility, endothelialization, burst pressure resistance. Among four types studied, namely, ePTFE, dePTFE, ePTFE‐P(AAm‐co‐NaAMPS)‐xanthan‐alginate, dePTFE‐P(AAm‐co‐NaAMPS)‐xanthan‐alginate, last graft exhibited most desirable attributes. It had highest longitudinal circumferential tensile strengths “toe region,” leakage pressures, suture retention strength. Additionally, also demonstrated best endothelialization capabilities. Overall, dePTFE‐P(AAm‐co‐NaAMPS)‐xanthan‐alginate found be promising candidate PTFE‐based grafts, showing potential future applications transplantation. Highlights Mechanical interlocking ensures robust bonding Hydrogel‐enhanced dePTFE‐hydrogel showed better capability.
