Lung cancer is the leading cause of death from in Ukraine and worldwide. The impact on tumor microenvironment most promising direction for lung therapy. Tumor-associated macrophages type M2 have powerful immunosuppressive properties. A new potential way influencing signaling protein activator transcription 6 (STAT6). aim our study was to evaluate role STAT6 formation prognosis patients with radically treated non-small cell (NSCLC). We performed an immunohistochemical examination tissue 42 NSCLC antibodies CD8+, forkhead box P3 (FOXP3+), CD163+, STAT6. survival assessed by Cox regression analysis. median follow-up period studied cohort 57.9 ± 4.2 months. 50.0% high expression established that correlates histology gender patients. High significantly more determined squamous carcinomas than adenocarcinomas. In carcinomas, metastasis regional lymph nodes associated immune exclusion phenotype mediated a infiltration stroma macrophages. An inflammatory low FOXP3+, typical adenocarcinoma. Low are common women; men. Females CD8+ clusters, CD163+ stroma, better overall survival. Conclusions: overexpression has negative recurrence-free To obtain accurate results, it necessary conduct includes larger patients, particular, female

ABSTRACT Bazhen Decoction (Eight Treasures Decoction) has demonstrated efficacy in the treatment of colorectal cancer (CRC), yet active ingredients it and mechanisms underlying their anti‐cancer properties are not well understood. Through network pharmacology, effective components against CRC corresponding key genes were delineated. Molecular docking was executed to identify component targeting gene CXCL8, which led discovery Quercetin. The cellular thermal shift assay method then used verify binding interaction. cells treated with incremental concentrations Quercetin, cell viability evaluated by Cell Counting Kit‐8 calculate IC 50 , apoptosis rates determined flow cytometry. Expression apoptosis‐related proteins Bcl‐2 Cleaved caspase‐3 measured using western blot. impact Quercetin on macrophage polarization studied co‐culturing macrophages, assessing M1 M2 distribution via cytometry, quantifying cytokine levels (IL‐6, IL‐10, IL‐12, CXCL8) enzyme‐linked immunosorbent (ELISA). ingredient from exhibited a targeted affinity enabled inhibit proliferation induce apoptosis. overexpression CXCL8 associated promotion malignancy, presence could lessen cells. Moreover, leads diminished abundance macrophages an increase cytokines IL‐6 while reducing IL‐10 indicates that inhibitory effect polarization. is known for anti‐CRC effects, targets inhibits impede malignant behaviors macrophages. Thus, may be utilized as immunomodulatory agent treatment.

M2-like tumor-associated macrophages (M2-like TAMs) function crucially in the tumor microenvironment (TME) and cancer development. This study developed a prognostic signature based on TAM-related genes for breast (BRCA) applying transcriptome scRNA-seq analysis. TCGA-BRCA, GSE20685, GSE176078 datasets were downloaded from UCSC xena GEO databases. AUCell score of immune-related (IRGs) was calculated using R package. Genes related to TAMs screened by WGCNA. Prognostic further identified univariate Cox LASSO regression analyses form RiskScore model, which validated external dataset. Furthermore, nomogram established integrating clinical characteristics, correlation analysis between TME or chemotherapeutic drugs conducted. Finally, mRNA expression levels key verified quantitative real time polymerase chain reaction (qRT-PCR). As exhibited highest IRGs single-cell transcriptomic atlas BRCA, cells classified into Macrophages C1 C2 subtypes, with subtype showing high M2 macrophage marker genes. ARHGAP26, RILP, KLRB1, CSTA, KLHDC7B, PSMB8, KYNU, RNASE1, LONRF3, TRPM2 as total 903 establish robust model. strong predictive performance constructed combining stage, Age, RiskScore, we found that most immune showed negative RiskScore. Multiple closely associated notably, Ribociclib_1632 had higher half-maximal inhibitory concentration (IC50) value high-risk group. qRT-PCR demonstrated 10 significantly different control BRCA cell lines. We providing potential therapeutic targets treatment cancer.

AGS-30, a new andrographolide derivative, showed significant anticancer and anti-angiogenic characteristics. However, its role in controlling macrophage polarization tumor immune response is unknown. Thus, the main goals of this study are to investigate how AGS-30 regulates it suppresses breast cancer metastasis. inhibited IL-4 IL-13-induced RAW 264.7 THP-1 macrophages into M2-like phenotype. did not affect LPS IFN-γ-induced M1-like macrophages. reduced mRNA expressions CD206, Arg-1, Fizz-1, Ym-1, VEGF, IL-10, MMP2, MMP9 concentration-dependent manner. In contrast, treatment at 5 μM polarization. The conditioned medium from increased 4T1 cell migration invasion, whereas these effects. xenograft mice, volume weight were without affecting body after receiving AGS-30. also lung liver metastasis, with STAT6, CD31, Ki67 protein expressions. Moreover, tumors had considerably fewer Arg-1 expression, but proportion iNOS expression treatment. Same results found tail vein metastasis model. conclusion, shows that inhibits growth probably through inhibiting Our findings suggest may be potential immunotherapeutic alternative for metastatic cancer.
