Artemisia campestris subsp. (tuguft) is a medicinal plant traditionally used in Algerian medicine. This study investigates the chemical composition and bioactivity of its essential oil (ACEO). Gas chromatography-mass spectrometry (GC-MS) analysis identified key compounds, including linalyl acetate (2.92%), geranyl (2.45%), eucalyptol (1.38%). ACEO demonstrated significant antioxidant activity, with IC50 values 11.09 μg/mL (DPPH), 15.81 (FRAP), 22.70 (β-carotene). It also enhanced peroxidase activity by 82.67 U/g. The anti-inflammatory effects were confirmed an 18.87 μg/mL. Notably, silico molecular docking revealed that 3-cyclopentyl-N-(2-(3,4-dimethoxyphenyl)ethyl) exhibits strong binding affinity to phosphoinositide 3-kinase gamma, target for pancreatic cancer therapy, suggesting potential anticancer activity. These findings underscore therapeutic ACEO, highlighting antioxidant, anti-inflammatory, properties.

Lead (Pb) is a common pollutant that not biodegradable and gravely endangers the environment human health. Annona squamosa fruit has wide range of medicinal uses owing to its phytochemical constituents. This study evaluated effect treatment with A. extract (ASFE) on testicular toxicity induced in male rats by lead acetate. The metal-chelating capacity composition ASFE were determined. LD50 was probit analysis. Molecular docking simulations performed using Auto Dock Vina. Forty Sprague Dawley equally divided into following groups: Gp1, negative control group; Gp2, given (350 mg/kg body weight (b. wt.)) (1/10 LD50); Gp3, acetate (PbAc) solution (100 b. wt.); Gp4, PbAc as Gp3 Gp2. All treatments oro-gastric intubation daily for 30 days. Body changes, spermatological parameters, reproductive hormone levels, oxidative stress inflammatory biomarkers evaluated, molecular histopathological investigations performed. results showed had promising activity composition. 3500 wt. analysis quercetin demonstrated high binding affinity JAK-1 STAT-3 proteins, this could make it more candidate targeting JAK-1/STAT-3 pathway than others. defective tissues, altered molecular, biochemical, histological features, well impaired characteristics. Treatment led significant mitigation these dysfunctions modulated JAK-1/STAT-3/SOCS-1 axis rats.

Squalene epoxidase (SQLE) is a pivotal enzyme in sterol biosynthesis, catalyzing the conversion of squalene to 2,3-oxidosqualene. Beyond its core role cholesterol homeostasis, SQLE implicated cancer, hypercholesterolemia, and fungal infections, positioning it as valuable therapeutic target. We conducted comprehensive literature search across primary databases gather vitro, silico, vivo evidence on SQLE. This review explores enzyme's structural functional features, including substrate specificity catalytic mechanisms, examines inhibitor interactions. Computational methods predict - dynamics, guiding drug design, while investigations clarify SQLE's metabolic disorders tumorigenesis. Challenges include resistance study discrepancies, but emerging technologies, such cryo-electron microscopy CRISPR editing, offer new avenues for deeper exploration. an underexplored yet promising target, with particular relevance oxidative stress, ferroptosis, gut microbiota research. Overcoming current barriers through advanced technologies multidisciplinary strategies could propel SQLE-targeted treatments into clinical practice, supporting precision medicine broader translational applications.

Mahuang Fuzi Xixin Decoction (MFX), a traditional Chinese medicine containing 44 volatile components (97.36% total oil), includes key compounds like α-terpenol (13.34%) and 1,2,3-trimethoxy-5-methyl-benzene (22.64%). Using network pharmacology Mendelian randomization, 24 active were identified targeting MS-related pathways (NF-κB, NLRP3, Toll-like receptor). Genetic variants in CYP450, GSK3B, CYBB, PON1 correlated with reduced MS risk. In EAE mice, MFX decreased spinal GSK-3B expression (immunofluorescence) pro-inflammatory factors (ELISA), demonstrating neuroprotection via anti-inflammatory, antioxidative mechanisms restored levels, highlighting therapeutic potential for MS.

Wistar rats were divided into three groups: negative control, positive control treated with indomethacin, group extract. Oxidative stress markers, cytokine levels, protein expressions assessed. demonstrated comparable those group. second study, effects assessed using tumor Ehrlich ascites carcinoma. Swiss albino mice four negative, cyclophosphamide (Cph), Group 3 Cph extract, 4 alone. Samples from fluid, liver, heart analyzed evaluate oxidative stress, inflammation, cancer markers. showed reduction tumor-associated inflammation stress. Overall, exhibited promising models studied. Background: This study aimed to examine the effect of 7-Ketositosterol (7-KSS), on sphingomyelin/ceramide metabolites and apoptosis in human breast MCF-7 liver HepG2 cancer cells. Methods: Anti-proliferative effects 7-KSS treatment were assessed at different concentrations periods. Cell viability was through MTT analysis, whereas levels sphingosine-1-phosphate (S1P), sphingomyelins (SMs), ceramides (CERs) measured using LC-MS/MS. Phosphorylated 44/42 ERK1/2 NF-κB p65 (Ser536) protein by Western blot analysis immunofluorescence staining. Apoptosis evaluated TUNEL staining flow cytometric assessment annexin-V propidium iodide (PI) labeling. Results: Treatment with significantly decreased cell survival S1P, p-44/42 ERK1/2, p-NF-κB cells compared controls. A substantial rise detected intracellular amounts C16-C24 CERs incubated 7-KSS. Conclusions: stimulated ceramide accumulation while decreasing proliferation via downregulating levels.
