Sperm motility is a key factor influencing male fertility and associated with metabolic lipid profiles across species. The aim of this study was to investigate the relationship between sperm seminal plasma profile in Simmental bulls, identify lipids potentially motility. Semen samples were collected from 26 healthy bulls an average age 4.9 years. quality evaluated using computer-assisted analysis (CASA). Based on motility, divided into two groups: high (HSM > 65%) low (LSM < 65%). Compared LSM group, HSM group exhibited significantly higher viability, straight-line velocity, beat-cross frequency, acrosome integrity, while malformation rate lower (p 0.05). Lipid determined LC-MS/MS, 40 differential identified by multivariate statistical analysis. Among them, 39 upregulated compared group. The protein proAKAP4 is crucial for sperm motility and has been suggested as an indicator of male fertility. We determined the relationship between concentration parameters in mice, investigated effects cryopreservation on these variables.Computer-assisted analysis ELISA were applied to determine fresh frozen-thawed epididymal SWISS, B6D2F1, C57BL/6N, BALB/c mice.ProAKAP4 levels ranged 12 89 ng/ml did not differ samples, or strains. found a negative some parameters. Abstract ProAKAP4 is gaining increasing attention as a potential marker of semen quality in many species, but while there commercial kit for assessing proAKAP4 the domestic cat, are no publications about its use. The aim this study was to evaluate – Cat 4MID® Kit (SPQI 4BioDx, Lille, France) assessment feline semen. Semen collected from 54 male cats by urethral catheterization. After basic evaluation (subjective motility, CASA, viability, morphology), levels each sample were assessed using according manufacturer's protocol or with some modifications related incubation time, storage conditions and number spermatozoa used. Finally, Spearman correlation concentration sperm motility parameters calculated. most reliable results (acceptable intraassay coefficient variation) obtained an optimized overnight isolation protein 1 × 10 6 stored at −80°C. For fresh semen, significant correlations between parameters, despite strong viability morphology. A predominant effect other highly variable performance lysis buffer question usefulness frozen–thawed moderate, negative two CASA VAP VSL. As cryopreserved samples, cannot be used freezability cats. More studies needed establish long‐lasting motility.
