Rheumatoid arthritis (RA) is a common chronic systemic autoimmune disease that often results in irreversible joint erosion and disability. Methotrexate (MTX) the first-line drug against RA, but significant side effects of long-term administration limit its use. Therefore, new therapeutic strategies are needed for treating RA. Here, dual-targeting biomimetic carrier-free nanomaterials (BSA-MTX-CyI nanosystem, BMC) developed synergistic photo-chemotherapy Bovine serum albumin (BSA), which has high affinity with SPARC (secreted protein acidic rich cysteine) RA microenvironment, selected as hydrophilic end coupled MTX phototherapeutic agent CyI to self-assemble into BMC. In vitro vivo experiments revealed BMC accumulated significantly at site collagen antibody-induced mice could be specifically recognized taken up by folate receptors proinflammatory M1 macrophages. Upon near-infrared laser irradiation, exerted photodynamic photothermal effects, whereas not only inhibited cell proliferation also increased sensitivity reactive oxygen species, enhancing apoptotic effect induced achieving photo-chemotherapy. Moreover, induce macrophages reprogram anti-inflammatory M2 This study provides innovative approaches treatment via macrophage apoptosis re-polarization.

Photodynamic therapy (PDT) involves the activation of photosensitizers (PSs) by visible laser light at target site to catalyze production reactive oxygen species, resulting in tumor cell death and blood vessel closure. The efficacy PDT depends on PSs, amount oxygen, intensity excitation laser. PSs have been extensively researched, great efforts made develop an ideal photosensitizer. Chlorin-e6 is FDA-approved second-generation that has attracted widespread research interest medical field, especially with respect antitumor anti-inflammatory activity. possesses advantages a large absorption coefficient, high strength, low residue body, relatively safety thus promising application prospects. Here we review use chlorin-e6 discuss prospects further development this technology.

Rheumatoid arthritis (RA) is an autoimmune disease whose pathophysiology closely related to inflammation-associated cells and the microenvironment of inflamed joints. This study aimed develop dual-targeted, reactive oxygen species (ROS)/pH dual-responsive, size-shrinkable intelligent micelles targeting M1 macrophages fibroblast-like synoviocytes (FLSs) enhance drug efficacy safety. These were surface-modified with PEG5000 prolong their circulation time in bloodstream hide molecules. The optimized particle size allowed reside joints through extravasation leaky vasculature subsequent inflammatory cell-mediated sequestration (ELVIS) effect. high concentration ROS joint caused detachment hydration layer PEG5000, which was then specifically recognized internalized by FLSs via CD44 receptor-mediated endocytosis, ultimately allowing release into acidic environment cells. vivo vitro evaluation showed that precisely targeted site, thus inhibiting expression pro-inflammatory cytokines, reversing polarization macrophages, invasion migration proliferative FLSs, and, at same time, regulating seeds soils RA. Rheumatoid Arthritis (RA) is a chronic inflammatory disease characterized by joint inflammation, progressive cartilage degradation, and bone erosion. Recent research has implicated ferroptosis not only in autoimmune hepatitis but also the pathogenesis progression of disorders like RA. Consequently, numerous therapeutic strategies have begun to target pathway, particularly design development nanodrug delivery systems (NDDSs). While previous reviews comprehensively discussed mechanisms ferroptosis, related signaling pathways, NDDS materials, recent studies further elucidated interplay between various metabolic providing robust theoretical basis for NDDS-based strategies. This review focuses on investigating role RA, aiming elucidate how targeting can offer novel concepts potential treatments RA patients. Specifically, it summarizes ferroptosis-based NDDSs via different pathways highlights feasibility treatment regimens based mechanism. Furthermore, critically discusses current limitations offers perspectives future directions this field.

Liposomes, as nanocarriers for therapeutics, are a prominent focus in translational medicine. Given their biocompatibility, liposomes suitable drug delivery systems rendering highly efficient therapeutic outcomes with minimal off-site toxicity. In different scenarios of human disease, it is essential not only to maintain levels but also target them the appropriate intracellular compartment. Mitochondria regulate cellular signalling, calcium balance, and energy production, playing crucial role various diseases. The notion focusing on mitochondria targeted was proposed several decades ago, yet practical application this idea its translation clinical use still development. Mitochondrial-targeted offer an alternative standard systems, potentially reducing off-target interactions, side effects, dosage or frequency. . To advance field, imperative integrate disciplines such chemical design, pharmacology, pharmaceutics, cell biology. This review summarises scientific advances development characterisation novel liposome-based targeting while revisiting potential.
