Abstract The number of people with hearing loss disorders is enormous, causing great physical and mental stress to patients, as well a huge social burden. Among these caused by inner ear lesions accounts for large proportion. Therefore, treatment the important. Inner drug delivery systems, which can reduce side effects systemic administration delivering drugs directly ear, are important in sensorineural loss. Here, development systems focused, including complex physiological structure that they face, types delivered, routes administration, forms carrier platforms. Recent studies this process presented it concluded summary outlook on problems faced possible solutions.

Abstract Severe to profound sensorineural hearing loss seriously affects the communication and cognitive ability of patients. Cochlear implantation (CI) is currently most effective treatment, while it may damage remaining inner ear function due its poor biocompatibility resultant fibrosis. Herein, a porous methacrylated poly(dimethylsiloxane) (MA‐PDMS)‐coated cochlear electrode presented for CI protection. The MA‐PDMS filled with hybrid hydrogel system made dexamethasone sodium phosphate (Dex), Ti 3 C 2 T x MXene (MXene), methacrylate gelatin (GelMA). coating shows good drug loading release capacity in vitro, protective effects on hair cells (HCs) spiral ganglion neurons (SGNs) ear, as well residual protection fibrosis reduction vivo. It anticipated that this drug‐loading provide valuable reference strategy future transplantation system.

Various non-invasive delivery systems have recently been developed as an alternative to conventional injections. Local transdermal administration represents the most attractive method due low systemic side effects, excellent ease of administration, and persistent drug release. The tympanic membrane (TM), a major barrier between outer middle ear, has similar structure stratum corneum compared surface skin. After several attempts, trans-tympanic regarded promising option in treatment inner ear diseases. round window (RWM) was possible approach from ear. improved permeability nanocarriers crossing RWM is current hotspot therapeutics for In this review, we include latest studies exploring treat Both passive active are described. A summary benefits disadvantages various clinical practice production procedures introduced. Finally, future approaches its effective application system characterised.
