Background and purpose: Cannabidiol (CBD) is a phenolic terpene compound with anticancer, antioxidant, anti-inflammatory, antibacterial, neuroprotective, anticonvulsant properties. Since the effects of CBD on sodium arsenite (As)-induced nephrotoxicity have not been fully determined, this study investigated effect As-induced by evaluating NOX4 NF-kB pathways in mice. Experimental approach: 48 male mice were divided into six groups (8 each) including group 1, receiving saline for 14 days; 2, (10 mg/kg, intraperitoneally) from 7 th to day; 3, As mg/kg) days gavage; treatment 4-6, (2.5, 5, 10 i.p.) 1.5 h before mg/kg gavage, days) day. Mice anesthetized after overnight fasting day 15, blood sample was collected their hearts. The level antioxidants pro-inflammatory factors, expression ROS TNF-α, NF-kB, NOX4, iNOS, cleaved PARP, caspase-3 proteins measured histological studies performed. Findings/Results: Exposure significantly increased kidney markers, oxidative stress, apoptosis, inflammation tissue, pretreatment reversed these changes. In addition, decreased levels factors PARP antioxidants. Conclusion implications: ameliorated related inhibiting inflammation, potentially through NF-kB/Nox4 pathway.

3,3',4,4',5-pentachlorobiphenyl (PCB126) is widely distributed, non-degradable and bioaccumulative, which can affect the function of tissues organs living organisms. Melatonin (MT) a sort indole neurohormone that mainly secreted by pineal gland. Numerous studies have shown MT alleviate intestinal injury through various mechanisms such as antioxidant, anti-inflammatory, anti-apoptosis. For above reasons, aim this study to explore mechanism in mice after exposure PCB126 well antagonistic effect MT. Mice were respectively fed (0.326 mg/kg) and/or (10 vivo. In vitro, colonic epithelial cells (MCEC) treated with (150 μM) (2 mM). We found microscopic structure colon tissue was impaired PCB126. The levels oxidative stress, protein mRNA expression inflammatory related factors significantly increased tight junction decreased. Notably, promote Nrf2/HO-1 level reduce caused Further vitro treatment reactive oxygen species inhibitors (NAC) showed it alleviated PCB126-induced MCEC cell damage. summary, results suggested alleviates inflammation inhibiting overproduction (ROS) up-regulating protein. Our contribute further comprehension toxicity effects significant role preserving injury.
