Abstract BACKGROUND Giant ragweed ( Ambrosia trifida L.) is one of the most troublesome weed species in corn Zea mays and soybean [ Glycine max (L.) Merr.] cropping systems. Herbicide-resistant weeds pose a threat to food production in modern agriculture, causing US$32 billion crop losses worldwide. In Michigan, highly troublesome and widespread include waterhemp, Palmer amaranth, common ragweed, horseweed, with accessions that are resistant glyphosate (Group 9) ALS-inhibitors 2), major herbicide sites of action utilized soybean corn cropping systems. Molecular assays for rapid resistance diagnostics confirm the in-field status can assist more effective, timely, proactive management. this research, we developed tested PCR-based identify target site mechanisms both groups through Sanger sequencing EPSPS copy number variation. Nine different SNPs were identified five ALS positions known confer among all species surveyed. Pro197Ser was most frequent horseweed ragweed accessions, whereas Trp574Leu predominant mutation amaranth waterhemp. Four contained Pro106Ser gene, which confers glyphosate. Additionally, waterhemp had 2–7 20–160 copies EPSPS, respectively. The validated by comparing genotyping several field-collected unknown susceptible accessions. efficacy >98% required only two days, confirming molecular robust tool diagnostics. Ambrosia trifida (giant ragweed) is an invasive species that causes habitat destruction and competitively excludes native plants in many parts of Europe Asia.
