This study was designed to investigate the protective effect and mechanism of Sufentanil on acute lung injury in septic mice based network pharmacology animal experiments, provide new ideas for clinical treatment. To this end, a protein-protein interaction (PPI) common targets first constructed with Swiss Target Prediction Database, GeneCards Draw Venn Diagram Software, STRING 11.5 Cytoscape 3.10.0 Software Metascape then key were subject enrichment analysis by Gene Ontology (GO) Kyoto Encyclopedia Genes Genomes (KEGG) obtain treatment pulmonary sepsis, verified experiments. A sepsis model cecal ligation puncture (CLP) study, tissues bronchoalveolar lavage fluid (BALF) taken from each group mice. The morphological changes apoptosis observed HE TUNEL staining, content inflammatory factors detected ELISA, expression proteins, such as p-JAK2 p-STAT3, Western blotting. According results pharmacology, total 40 screened out GO involved 1,483 biological processes (BPs), 84 cellular components (CCs) 125 molecular functions (MFs); KEGG identified 137 signaling pathways p < 0.05 JAK-STAT. compared control group, had severe tissue elevated relevant tissue. Compared CLP + showed reduced pathomorphologic lesions, lower level, well p-STAT3 proteins experiments consistent pharmacology. In summary, may improve inhibiting JAK2-STAT3 pathway, which provides basis research

Abstract Purpose Ubiquitination plays a crucial role in various diseases. This study aims to explore the potential ubiquitination related genes IPF. Methods The gene microarray dataset GSE24206 was obtained from GEO database. Subsequently, through differential expression analysis and molecular signatures database, we 1734 differentially expressed 742 genes. Through venn diagram analysis, 53 Then, gene-ontology (GO) enrichment Kyoto Encyclopedia of Genes Genomes (KEGG) pathway protein-protein interactions (PPI) set (GSEA) were applied for Finally, CDC20 ITCH IPF patients cells validated by qPCR western blot assay. Results A total (36 up-regulated 17 down-regulated genes) identified between 6 healthy controls. GO KEGG mainly involved regulation protein ubiquitination, post-translational modification ubiquitin mediated proteolysis. PPI results demonstrated that these interacted with each other. GSEA some hub epithelial mesenchymal transition, inflammatory response, hypoxia, apoptosis. experiment level consistent bioinformatics results. Conclusion We analysis. other may influence development transition response. Our research findings provide insights into mechanisms fibrosis evidence therapeutic targets fibrosis.

Sepsis, a severe condition instigated by infections, continues to be primary global cause of death, typified systemic inflammation and advancing immune dysfunction. Comprehending the complex pathological processes that underlie sepsis is integral creation efficacious treatments. Despite inability animal models entirely reproduce clinical intricacies related sepsis, they are invaluable instruments for exploration development therapeutic approaches. Within this context, zebrafish model particularly noteworthy due its genetic tractability, transparency, appropriateness high-throughput screening mutants compounds. This scholarly review emphasizes crucial role disease plays in enhancing our comprehension exploring applications deciphering inflammatory responses, evaluating consequences alterations, examining novel agents. The Insights derived from research not only augment understanding underlying mechanisms but also possess considerable potential transference these discoveries into therapies, thus potentially transforming approach management. objective article underscore importance realm biomedical pertaining delineate forthcoming opportunities utilizing applications.
