Cultivated potatoes are incredibly diverse, ranging from diploid to pentaploid and encompass four different species. They adapted disparate environments conditions carry unique alleles for resistance pests pathogens. Describing how diversity is partitioned within among these populations essential understanding the potato genome effectively utilizing landraces in breeding. This task complicated by difficulty of making comparisons across cytotypes extensive admixture section petota. We genotyped 730 accessions US Potato genebank including wild diploids cultivated tetraploids using Genotype-by-sequencing. data set allowed us interrogate population structure as well generate core subsets which will support breeders efficiently screening material biotic abiotic stress alleles. found that even controlling ploidy, tetraploid exhibited higher observed expected heterozygosity than accessions. In particular group chilotanum was most heterozygous only taxa not exhibit any inbreeding. may part be because has a history introgression just species, but well. Solanum section Petota Dumort. consists of tuber-bearing species (i.e., the cultivated potatoes and their wild relatives) that have both asexual sexual propagation, variation in ploidy, reproductive isolation. These undergone adaptation to a diversity climates, altitudes, photoperiods, geographical range. The defies characterization with biological concept due interspecies hybridization, allo- auto-polyploidy, phenotypic plasticity. Genetic studies, more recently genome sequencing pangenome analyses, are fostering greater understanding genetic processes shape evolution speciation section, shedding light on phylogeny providing insights utilization potato crop relatives breeding for climate-resilient varieties.

We genotyped a population of 618 diploid potato clones derived from six independent potato-breeding programmes NW-Europe. The diploids were phenotyped for 23 traits, using standardized protocols and common check varieties, enabling us to derive whole estimators most traits. subsequently performed genome-wide association study (GWAS) identify quantitative trait loci (QTL) all traits with SNPs short-read haplotypes read-backed phasing. In this study, we used marker platform called PotatoMASH (Potato Multi-Allele Scanning Haplotags); pooled multiplex amplicon sequencing based approach. Through method, neighboring within an can be combined generate multiallelic (haplotags) that capture recombination history between the constituent reflect allelic diversity given locus in different way than single bi-allelic SNPs. found total 37 unique QTL across both types. A core 10 was detected as well haplotags. Haplotags allowed detect additional 14 not on SNP set. Conversely, set also 13 detectable haplotag conclude types should routinely parallel maximize detection power. report 19 novel nine traits: Skin Smoothness, Sprout Dormancy, Total Tuber Number, Length, Yield, Chipping Color, After-cooking Blackening, Cooking Type, Eye depth.
