This review describes our current understanding of the role mitochondria in repurposing anti-diabetes drugs metformin, gliclazide, GLP-1 receptor agonists, and SGLT2 inhibitors for additional clinical benefits regarding unhealthy aging, long COVID, mental neurogenerative disorders, obesity. Metformin, most prominent these diabetes drugs, has been called “Drug Miracles Wonders,” as trials have found it to be beneficial human patients suffering from maladies. To promote viral replication all infected cells, SARS-CoV-2 stimulates liver cells produce glucose export into blood stream, which can cause COVID patients, reduces levels blood, was shown cut incidence rate half recovering SARS-CoV-2. Metformin leads phosphorylation AMP-activated protein kinase AMPK, accelerates import via transporter GLUT4 switches starvation mode, counteracting virus. Diabetes also stimulate unfolded response thus mitophagy, is healthy aging health. were mimic exercise help reduce body weight.

Abstract Fibromyalgia (FM) patients are highly susceptible to depression. Wnt/β-catenin signaling has shown a crucial role against depression in several studies. The FDA-approved FM drug, milnacipran (Miln), antinociceptive potential FM. Yet, no study investigated its antidepressant Vanillin (Van), well-known phytochemical often employed as flavoring agent, been previously reported for and effects animal models, but not tested so far This explored the effect of Van Miln through investigating signaling. was induced female Wistar rats by injecting reserpine (1 mg/kg/day s.c) 3 days. Thereafter, animals received either (30 p.o) or (100 subsequent 14 Results showed that both drugs demonstrated forced swimming test besides analgesic, antiallodynic influences observed Randall-Selitto, hot plate, cold allodynia, Von-Frey, tail immersion tests. Biochemically, significantly enhanced serotonergic transmission hippocampus upregulated protein expression Wnt/GSK-3β/β-catenin axis, including downstream proteins, T cell factor, dicer. is followed upregulation resilience micro ribonucleic acids (miRNAs) 124 135. Histopathological examinations corroborated biochemical molecular findings. Interestingly, these were overturned via administration β-catenin inhibitor, XAV939 (0.1 mg/kg, i.p., daily). In conclusion, this outlined aptitude activating reserpine-induced

Recently, a novel concept of 'anti-inflammatory-based pharmacotherapy' has been developed. The aim this scoping review was precisely to explore in depth the current status inflammation randomised controlled clinical trial studies depression. PubMed systematically searched from inception December 11 2024. Studies that researches on treatment depression based anti-inflammatory strategies were included. Study characteristics and outcomes extracted organized thematically. reports trials included, which accumulated 119 depressed patients. All found there is connection between effects drugs treating large decrease levels inflammatory markers blood patients compared before treatment. Three markers, CRP, IL-6 TNF-alpha, most frequently mentioned. strategy drug administration did not differ fundamentally previous traditional antidepressant combined with psychotherapy. Abstract One of the major consequences diabetes mellitus that has gained attention due to its rising incidence is cognitive impairment. Recent research suggested sodium-glucose cotransporter-2 (SGLT-2) inhibitors can mitigate memory impairment linked Alzheimer’s disease and are now being explored for their benefits. However, mechanisms were not thoroughly studied. This investigates hypothesis neuroprotective effect empagliflozin administration against scopolamine-heavy metal mixture (SCO + HMM)-treated rat models in comparison with memantine as a reference drug impact combination. Yet, effects combination have been previously explored. study employed adult male albino rats categorized into five groups. The empagliflozin, memantine, concomitant on performance was assessed scopolamine heavy mixture-treated model rats. assessment rats’ behavior, memory, spatial learning conducted, followed by an evaluation hippocampal brain-derived neurotrophic factor (BDNF), beta-secretase (BACE-1), oxidative stress (OS), inflammatory marker activity. And, western blot analysis conducted detect phosphorylated 5’ AMP-activated protein kinase (p-AMPK), mammalian target rapamycin (p-mTOR), heme oxygenase-1 (HO-1). Hippocampal cerebellar histopathology examined, addition expressions amyloid β (Aβ). current data demonstrate involvement pAMPK/mTOR/HO-1 signaling pathway neuroprotection SCO HMM-induced AD. In addition, it reduces AD hallmarks (Aβ BACE1), neuro-inflammation, sequelae, enhances neurogenesis synaptic density via BDNF. proposes EMPA, especially when co-administered other conventional anti-Alzheimer therapy, may be formulated innovative therapeutic strategy enhancement impairments associated neurodegenerative disorders.
