There has been limited research into arsenolipid toxicological risks and health-related outcomes due to challenges with their separation, identification, quantification within complex biological matrices (e.g., fish, seaweed). Analytical approaches for identification such as suspect screening have not well documented there are no certified standard reference materials, leading issues reproducibility uncertainty regarding the accuracy of results. In this study, a detailed workflow arsenolipids utilizing coupled data independent analysis is presented applied three commercially available materials (Hijiki seaweed, dogfish liver, tuna). Hexane dichloromethane/methanol extraction, followed by reversed-phase high-performance liquid chromatography-inductively plasma mass spectrometry chromatography-electrospray ionization-quadrupole time-of-flight spectrometry. Using developed, fragmentation matching, error calculations, retention time matching were performed identify arsenolipids. Arseno-fatty acids (AsFAs), arsenohydrocarbons (AsHCs), arsenosugar phospholipids (AsSugPLs) identified high confidence; AsHC332, AsHC360, AsSugPL720 in AsHC332 tuna, AsFA474 AsFA502 liver. promising candidates further work on synthesis, using MS/MS, toxicity testing.
