ABSTRACT 1-Deoxy-D-xylulose-5-phosphate synthase (DXS) is the first key enzyme in MEP responsible for monoterpene synthesis. To reveal its regulatory effect on floral scents of lily, LiDXS2 was cloned from Oriental Lily ‘Sorbonne’; protein location determined using subcellular localisation; and expression pattern different flowering stages tissues probed by fluorescence quantitative PCR. The plant vector constructed to transform Arabidopsis thaliana , transgenic plants were characterised. results showed that ORF area 2142 bp encoded a total 713 amino acids. Analysis homologous sequences it contains two highly conserved regions, WDVGHQ IAEQHA, which closely related Ginkgo DXS2 Periwinkle DXS protein. Subcellular localisation gene Lilium 'Sorbonne' localised chloroplasts. In stages, patterns basically same as TranScriptome sequencing results, overall trend initially increasing then decreasing. organ tissues, level significantly higher petals than other almost no observed anthers. A. transformed lines an increased height early phenotype, volatile synthesis genes structures downstream pathway upregulated compared with controls. Collectively, these suggest may play important role regulating ‘Sorbonne’ flavour release.
