Sucrose phosphate synthase (SPS) is an important enzyme in the sucrose synthesis of plants, governing accumulation and distribution photosynthetic assimilates, which essential for plant growth stress tolerance. This study successfully identified eight NtSPS genes within genome cultivated tobacco. Phylogenetic analysis revealed that these are categorized into three subfamilies, a classification supported by examination their gene structures conserved domains. The promoters NtSPSs contained variety cis-elements associated with development, responses to phytohormones, resistance. Expression profiling demonstrated exhibit distinct expression patterns across different tissues under various conditions. Notably, majority genes, especially NtSPS5 NtSPS6, showed high leaves increased both roots following drought treatment. Furthermore, overexpression NtSPS6 tobacco plants significantly improved germination rate mannitol treatment enhanced activity antioxidant enzymes along chlorophyll fluorescence parameters stress. These results suggest have positive impact on tolerance plants. Therefore, this provides significant target resistance breeding lays foundation further investigation function regulatory mechanisms SPS genes.
