Comammox, as a newly discovered ammonia oxidizer, urgently needs highly efficient and specific primers to detect its community structure diversity. In this study, the performance of widely used primer set Ntsp-amoA 162F/359R designed sets comamoA F/R, CA377f/C576r, CB377f/C576r were evaluated, for high-throughput sequencing comammox amoA genes in natural arable soils sampled from two locations black soil region northeast China. Results showed that, compared with had more advantages detecting operational taxonomic unit (OTU) numbers, diversity, structure. The CA377f/C576r an advantage sequences low relative abundance. addition, results phylogenetic tree abundance dominant OTUs that China was dominated by Nitrospira Clade B. Furthermore, our study found long-term land use reduced alpha diversity community, but lead convergent evolution Mantel test canonical correspondence analysis indicated NO3--N content most important factor affecting comammox.
