Abstract Stemness, giving cancer cells massive plasticity enabling them to survive in dynamic (e.g. hypoxic) environments and become resistant treatment, especially chemotherapy, is an important property of aggressive tumours. Here, we review some essentials stemness focusing on triple-negative breast (TNBC), the most form all cancers. TNBC express a range genes mechanisms associated with stemness, including fundamental four “Yamanaka factors”. Most evidence concerns transcription factor / oncogene c-Myc interesting case expression neonatal splice variant voltage-gated sodium channel subtype Nav1.5. On whole, measures that reduce make less aggressive, reducing their invasive/metastatic potential increasing/restoring chemosensitivity. Such include gene silencing techniques, epigenetic therapies as well novel approaches like optogenetics aiming modulate plasma membrane voltage. Indeed, simply hyperpolarizing can stem differentiate. Finally, give overview clinical aspects exploitation cancer/TNBC diagnostics therapeutics. In particular, personalised mRNA-based mechanistically meaningful combinations are promising emerging discipline ‘cancer neuroscience’ providing insights both issues applications. Graphical
