Abstract Major depressive disorder (MDD) is a psychiatric disease of still poorly understood molecular etiology. Extensive studies at different levels point to high complexity numerous interrelated pathways as the underpinnings depression. systems under consideration include monoamines, stress, neurotrophins and neurogenesis, excitatory inhibitory neurotransmission, mitochondrial dysfunction, (epi)genetics, inflammation, opioid system, myelination, gut-brain axis, among others. This review aims illustrating how these multiple signaling may interact provide more comprehensive view MDD’s neurobiology. In particular, considering pattern synaptic activity closest physical representation mood, emotion, conscience we can conceptualize, each pathway or system will be scrutinized for links neurotransmission. Models neurobiology MDD discussed well future actions improve understanding treatment options.

Depression is one of the most fatal mental diseases, and there currently a lack efficient drugs for treatment depression. Emerging evidence has indicated oxidative stress as key pathological feature We targeted reactive oxygen species (ROS) synthesized CeO2@BSA nanoclusters novel antidepression nanodrug via convenient, green, highly effective bovine serum albumin (BSA) incubation strategy. ultrasmall size (2 nm) with outstanding ROS scavenging blood-brain barrier crossing capacity, rapid metabolism, negligible adverse effects in vitro vivo. administration alleviates depressive behaviors depression-related changes chronic restraint stress-induced model, suggesting promising therapeutic Our study proved validity by directly using nanodrugs instead them nanocarrier, which greatly expands application nanomaterials depression treatment.

Excessive fructose intake is associated with the increased risk of mental illness, such as depression, but underlying mechanisms are poorly understood. Our previous study found that high diet (FruD)-fed mice exhibited neuroinflammation, hippocampal neurogenesis decline and blood-brain barrier (BBB) damage, accompanied by reduction gut microbiome-derived short-chain fatty acids (SCFAs). Here, we chronic stress aggravated these pathological changes promoted development depressive-like behaviors in FruD mice. In detail, decreased number newborn neurons, mature neurons neural stem cells (NSCs) hippocampus was worsened stress. Furthermore, exacerbated damage BBB integrity expression zonula occludens-1 (ZO-1), claudin-5 occludin brain vasculature, overactivated microglia neuroinflammation These results suggest combined leads to cumulative negative effects promote Of note, SCFAs could rescue decline, improve suppress activation thereby ameliorate exposed be used develop dietary interventions prevent depression.

The gut microbiota influences host brain function, but the underlying gut-brain axis connections and molecular processes remain unclear. One pathway along this bidirectional communication system involves circulating microbially derived metabolites, such as short-chain fatty acids (SCFAs), which include butyrate propionate. Brain endothelium is main interface of between signals brain, it constitutes component blood-brain barrier (BBB). Here, we used a well-established in vitro BBB model treated with physiologically relevant concentrations propionate without lipopolysaccharide (LPS) to examine effects SCFAs on actin cytoskeleton tight junction protein structure. Both induced distinct alterations filamentous directionality. also increased spikes protected from LPS-induced tight-junction mis-localization, improved integrity, modulated mitochondrial network dynamics. These findings identify cytoskeletal dynamics another target further illuminating how can influence physiology.

The blood brain barrier (BBB) is a vital structure to protect the brain, tightly filtering passage of nutrients and molecules from brain. This critical for maintaining proper functioning any disruption in BBB has detrimental consequences often leading diseases. It not clear whether occurs first depression or consequence disease, however been observed depressed patients evidence points role important culprits depression, stress inflammation disrupting integrity BBB. mechanisms whereby stress, affect remain be fully understood. Yet, cytokines regulating tight junction protein expression seems crucial. Altogether, findings suggest that acting at level might provide therapeutic benefit depression.

Emerging evidence suggests that an important function of the sleeping brain is removal wastes and toxins from central nervous system (CNS) due to activation waste (BWRS). The meningeal lymphatic vessels (MLVs) are part BWRS. A decrease in MLV associated with Alzheimer’s Parkinson’s diseases, intracranial hemorrhages, tumors trauma. Since BWRS activated during sleep, a new idea now being actively discussed scientific community: night stimulation might be innovative promising strategy for neurorehabilitation medicine. This review highlights trends photobiomodulation BWRS/MLVs deep sleep as breakthrough technology effective unnecessary compounds order increase neuroprotection CNS well prevent or delay various diseases.
