Evaluating key traits of male sterile mutant accessions in rhizomatous alfalfa (Medicago sativa L.) is crucial for selecting plants artificial hybrid breeding maternal lines. In this study, branch cuttings from four ‘Qingshui’ were used as experimental samples. We evaluated phenotypic traits, which included pollen viability and stigma receptivity, well nutritional quality, using difference analysis, correlation principal component analysis. Prioritizing while considering quality supplementary factors, allowed us to comprehensively evaluate 24 individuals. This evaluation led the identification with superior traits. The accession 4-4 was found be partially fertile, whereas remaining 23 individuals entirely sterile. All exhibited receptivity levels suitable effective pollination. Principal analysis revealed that among assessed leaf–stem ratio contributed most significantly, followed by crude protein content, neutral detergent fiber content had least impact on overall quality. Additionally, number branches showed a strong positive individual plant yield (p < 0.01). No significant correlations detected height, stem diameter, forage grading index, protein, fiber, acid yield. Overall, our comprehensive suggests 1-2, 2-2, 3-1, 4-3 are use parental lines breeding.
