The chromosome-level genome assembly of Citrullus colocynthis reveals its genetic potential for enhancing drought tolerance, paving the way innovative crop improvement strategies. This study presents first comprehensive and annotation colocynthis, a drought-tolerant wild close relative cultivated watermelon, highlighting agricultural resilience to climate change. achieved using advanced sequencing technologies, including PacBio HiFi Hi-C, revealing size approximately 366 Mb with low heterozygosity substantial repetitive content. Our analysis identified 23,327 gene models, that could encode stress response mechanisms species' adaptation arid environments. Comparative genomics closely related species illuminated evolutionary dynamics within Cucurbitaceae family. In addition, resequencing 27 accessions from United Arab Emirates (UAE) diversity, suggesting foundation future breeding programs. genomic resource opens new avenues de novo domestication C. offering blueprint developing crops enhanced disease resistance, nutritional profiles, crucial sustaining food security in face escalating challenges.
