Oocyte aging is a key constraint on oocyte quality, leading to fertilization failure and abnormal embryonic development. In addition, it likely generate unfavorable assisted reproductive technology (ART) outcomes. SCM-198, synthetic form of leonurine, was found rescue the rate fragmentation caused by postovulatory aging. Therefore, aim this study conduct more in-depth investigation SCM-198 exploring its relationship with aged oocytes after ovulation or maternal clarifying whether affects cell quality. The results indicate that, compared group, 50 µM group significantly improved sperm-egg binding increased oocytes, restoring spindle apparatus/chromosome structure, cortical granule distribution, ovastacin Juno protein distribution. showed normal mitochondrial low levels reactive oxygen species (ROS), small quantity early apoptosis group. Above all, in vivo supplementation effectively eliminated excess ROS reduced spindle/chromosome structural defects mouse oocytes. summary, these findings that inhibits excessive oxidative stress alters quality both vitro vivo.

In mammals, oocytes display compromised quality after experiencing a process of postovulatory aging. However, the mechanisms underlying are not yet fully understood. Here, we portrayed protein expression profile fresh and aging metaphase II (MII) mouse by means four-dimensional label-free quantification mass spectrometry (4D-LFQ).The analysis 4D-LFQ data illustrated that there were seventy-six differentially expressed proteins (DEPs) between two groups MII stage oocytes. Fifty-three DEPs up-regulated while twenty-three down-regulated in group, Gene Ontology (GO) revealed these mainly enriched regulation gene expression, biosynthesis, RNA metabolism cell cycle. Our detailed related to processes such as transcription, translation, post-translational modifications epigenome was changed; relative metabolic processes, alternative splicing, export from nucleus negative transcription polymerase promoter also altered.In conclusion, identified considerable discussed how they agreed with previous researches illustrating altered associated research provided new perspective on established theoretical support for practical methods control reverse
