Metabolic syndromes (MetS) are complex metabolic disorders, the pathogenesis of which has not been fully elucidated. In recent years, association between gut microbiota and MetS attracted widespread attention, but causal relationship remains unclear. We performed a 2-sample Mendelian randomization analysis (MR) to examine whether is causally related its components find basis for potential diagnostic or intervention approaches MetS. utilized summary statistics from whole-genome analyses MiBioGen consortium obtained MetS-related data UK Biobank, IEU Open GWAS project, The Meta-Analyses Glucose Insulin-related traits Consortium (MAGIC). MR were using inverse variance weighted, MR-Egger, weighted median. Sensitivity conducted verify robustness results. Among 211 microbiota, we identified 8 that significantly associated with risk Specifically, Lachnospiraceae (family), Veillonellaceae Victivallaceae Odoribacter (genus), Olsenella (genus) may increase MetS, while Bifidobacteriaceae Ruminococcaceae UCG-010 Actinobacteria (phylum) decrease Additionally, discovered multiple various such as BMI, hypertension, blood lipid levels. This study first use methods reveal specific providing new perspective understanding offering important evidence development microbiota-based prevention treatment strategies

Abstract Background Most forms of obesity are associated with chronic diseases that remain a global public health challenge. Aims Despite significant advancements in understanding its pathophysiology, effective management is hindered by the persistence knowledge gaps epidemiology, phenotypic heterogeneity and policy implementation. Materials Methods This consensus statement European Society for Clinical Investigation identifies eight critical areas requiring urgent attention. Key include insufficient long‐term data on trends, inadequacy body mass index (BMI) as sole diagnostic measure, recognition diversity obesity‐related cardiometabolic risks. Moreover, socio‐economic drivers transition across phenotypes poorly understood. Results The syndemic nature obesity, exacerbated globalization environmental changes, necessitates holistic approach integrating frameworks community‐level interventions. advocates leveraging emerging technologies, such artificial intelligence, to refine predictive models address variability. It underscores importance collaborative efforts among scientists, policymakers, stakeholders create tailored interventions enduring policies. Discussion highlights need harmonizing anthropometric biochemical markers, fostering inclusive narratives combating stigma obesity. By addressing these gaps, this initiative aims advance research, improve prevention strategies optimize care delivery people living Conclusion effort marks decisive step towards mitigating epidemic profound impact systems. Ultimately, should be considered being largely consequence model not compatible optimal human health.

Objective . The importance of early microbial dysbiosis in later development obesity and metabolic disorders has been a subject debate. Here we tested cause effect mice. Methods Germ‐free male Swiss Webster mice were colonized specific‐pathogen‐free (SPF) facility at 1 week (1W) 3 weeks (3W) age. They challenged with high‐fat diet their responses compared SPF Gut microbiota was analyzed by 16S rRNA gene sequencing. Moreover, RNA sequencing the liver performed on additional 3W regular chow diet. Results There no significant differences weight, food consumption, epididymal fat HbA1c levels, serum insulin leptin, whereas germ‐free period resulted impaired glucose tolerance. Both 1W group peaked 56% ( p < 0.05) 66% 0.01) higher blood than control group, respectively. This accompanied 45% reduction level anti‐inflammatory cytokine IL‐10 0.05). gut between groups, indicating that all fully after period. Marked effects hepatic expression (728 differentially expressed genes adjusted 0.05 fold change ± 1.5) suggested potential predisposition to risk developing resistance group. Conclusions Lack microbes life had impact adiposity but led altered related metabolism study strongly supports notion signaling beginning can alter host’s disorder life.

Bacterial microbiota in stool may vary over a wide range, depending on age, nutrition, etc. The purpose of our work was to discriminate phyla and genera intestinal bacteria their biodiversity within healthy population (North-Western Russia) compared the patients with type 1 diabetes mellitus (T1DM). study group included 183 persons 2 53 years old (a mean 26.5±1.0 old), 41 T1DM (mean age 18.2±1.8 old). disease onset at 11±1.5 years, experience 7±1.5 years. Total DNA isolated from samples, sequencing libraries were prepared by amplifying V3-V4 region 16S rRNA gene sequenced Illumina MiSeq. Bioinformatic processing NGS databases adapted for evalutaion. Despite broad scatter, biological diversity bacterial expressed as Shannon index significantly increased younger older ages comparison group, higher adult persons, trend decrease Actinomycetota phylum which includes Bifidobacterium longum species. Similar but non-significant trends noted group. Concordant Bacillota prevalence samples diabetic patients, some anaerobic (Faecalibacteria, Lachnospira Ruminococcae, Roseburia) enriched microbiome against controls. Hence, correction Ruminococcus Lachnospiraceae requires future search new probiotics. Lower abundance Bifidobacter suggests potential usage Bifidobacter-based probiotics this cohort.

Sarcopenia is a syndrome with age-related muscle mass loss and physical function decline, which increases the risk of falls, frailty, malnutrition, hospitalization death. This study aimed to investigate incidence influencing factors inpatients sarcopenia, provided evidence for early prevention management sarcopenia. Methods PubMed, EMbase, The Cochrane library, Web Science, CNKI, VIP, WanFang, CBM databases were searched from establishment database December 2023, we use Stata14 software meta-analysis. Results A total 19,228 patients included in 38 literatures, including 4,766 results meta-analysis showed that prevalence sarcopenia was 32.9% (95%CI: 28.2%~37.5%), subgroup analysis highest Australia (71%) lowest Korea (12%). Grouped by gender, 50% 49%~52%) males 48% 47%~50%) females. Meta-analysis age, diabetes, mulnutrition, MNA-score, long stay hospital, hyperlipemia, dyskinesia, dementia, osteoporosis, oral feeding, cancer BMI (OR=0.78, 95%CI: 0.638~0.95), calf circunference (OR=0.92, 0.78~1.06) protective factor Conclusion higher than female. are complex, gender other cannot be changed. However, hyperlipidemia can changed, clinical works need establish prediction models according different disease types actively intervene high-risk as soon possible reduce occurrence diseases improve quality life patients.
