Due to the spread of antimicrobial resistance, there is an urgent need search for novel agents. In poultry farming, animals are constantly exposed enteral pathogens and simultaneously required maintain high production performance, making it challenging preserve homeostasis. Host defense peptides (HDP), regarded as putative antibiotic substitutes, have been considered beneficial gut health; however, many yet be investigated from this perspective. present study, chicken ileal explant cultures were used examine effects HDP Pap12-6 (PAP) on immune state tight junction (TJ) protein abundance. The peptide was applied at 25 50 µg/ml concentrations alone (PAP-low PAP-high, respectively) in inflammatory conditions evoked by polyinosinic-polycytidylic acid (Poly I:C, µg/ml), same doses (PI:C+PAP-low PI:C+PAP-high, respectively). According results, PAP did not influence metabolic activity extracellular lactate dehydrogenase activity, hence viability explants. Regarding state, level interleukin (IL)-2 increased PAP-low, PI:C+PAP-low, whereas IL-8 concentration raised PI:C+PAP-low treatment. Furthermore, PI:C+PAP-high decreased elevated interferon (IFN)-γ caused Poly I:C. case TJ proteins, PAP-low expression occludin, PAP-high abundance claudin-3. Conclusively, displayed immunomodulatory may contributed epithelial integrity without being cytotoxic, suggesting its potential alternative farming. However, detailed mechanism action concentration-dependent must elucidated future.
