Introduction Natural compounds have emerged as promising candidates in drug development due to their potent immunomodulatory anti-inflammatory, antibacterial, analgesic, and healing properties. They shown significant therapeutic potential clinical applications, such mouth rinses, toothpastes, localized delivery systems. The use of natural alternatives can contribute tackling antimicrobial resistance. Among compounds, curcumin has gained particular attention, demonstrating robust anti-cancer, antibiotic, anti-inflammatory activities numerous vivo studies, while exhibiting a favorable safety profile for the treatment various diseases. In this study, remedial effects its metabolite, tetrahydrocurcumin, on dental pulp were explored. addition, these results compared with our previous findings periodontal ligament gingival epithelial cells, further broadening understanding oral disease caries periodontitis. Methods RNA sequencing was used investigate differentially expressed genes cells following treatments tetrahydrocurcumin. Results We show that 1 μM or tetrahydrocurcumin is sufficient promote Wnt signaling pathway cells. Curcumin promotes upregulation cellular metabolism enhances response stress. Our enrichment analysis shows modulates extracellular matrix angiogenesis. Conclusions study highlight cytoprotective regenerative properties These could be leveraged approach tissue regeneration
