Background Obesity is associated with insulin resistance (IR) and metabolic dysfunction-associated steatotic liver disease (MASLD). Genistein, an isoflavone, a promising natural compound for preventing treating obesity dysfunctions. We aimed to investigate the sex-specific protective effects of genistein on obesity, IR, MASLD in murine model sex hormone deprivation diet-induced (DIO), mimicking postmenopausal women or aging men syndrome. Methods Gonadectomized sham-operated C57BL/6NJcl mice were fed high-fat high-sucrose diet 4 weeks induce (7 per group). In gonadectomized mice, (16 mg/kg/day) vehicle (7.5% dimethyl sulfoxide) was orally administered 45 days. assessed glucose homeostasis parameters, hepatic histopathology, gene expression gonadectomy treatment. Results Gonadectomy exacerbated adiposity both sexes. Ovariectomy diminished female gonadal hormones homeostatic assessment (HOMA-IR), serum alanine transaminase levels, steatosis score, genes progression such as Fasn , Srebf1 Saa1 Cd36 Col1a1 Pck1 Ppargc1a . Genistein treatment significantly reduced body weight gain score However, attenuated HOMA-IR only mice. Conclusion mitigates DIO-related male Regarding beneficial effect evident This study suggests potential alternative application individuals deprivation, yet pending clinical trials.
