Abstract Background Perioperative neurocognitive disorder (PND) is a prevalent form of cognitive impairment in elderly patients following anesthesia and surgery. The underlying mechanisms PND are closely related to perineuronal nets (PNNs). PNNs, which complexes extracellular matrix primarily surrounding neurons the hippocampus, play critical role function. Connexin 43 (Cx43) contributes function by modulating components PNNs. This study was designed investigate specific regulatory Cx43 on PNNs its pivotal development PND. Methods Eighteen-month-old wild-type Gja1fl/fl C57BL/6 mice were subjected abdominal surgery under 1.4% isoflurane anesthesia. Cognitive functions, particularly learning memory, evaluated via Y-maze test, Barnes maze (BM) contextual fear conditioning test (CFT). mRNA protein expression levels assessed using quantitative reverse transcription polymerase chain reaction (qRT-PCR), fluorescent situ hybridization (FISH), western blotting flow cytometry. quantity measured Wisteria floribunda agglutinin (WFA) Aggrecan staining. Results Aged exhibited deficits hippocampus-dependent accompanied increased expression. Conditional knockout (cKO) astrocytes alleviated promoted number dendritic spines hippocampus targeting Dmp1. Knockdown Dmp1 attenuated beneficial effects cKO induced Conclusion Our findings indicate that induce an increase expression, inhibits formation suppressing transcription, leading aged mice. These results offer new mechanistic insights into pathogenesis identify potential targets for therapeutic intervention.

Regular physical activity has been shown to ameliorate cognitive decline associated with aging, and improve autonomic nervous function. However, the effect of active housework, a form light moderate activity, on postoperative function in elderly remain unknown. The aim this study is investigate whether housework exerts protective against delayed neurocognitive recovery (dNCR), explore role vagal relationship. This prospective nested case-control that enrolled 152 patients scheduled for elective knee replacement surgery. neuropsychological assessments were conducted evaluate dNCR both preoperatively one week postoperatively. Physical levels quantified based Activity Scale Elderly categorized into sedentary behavior groups using hierarchical clustering. High-frequency spectral analysis heart rate variability was used assess activity. Multivariable logistic regression employed examine association between dNCR.
