Fungal biota represents important constituents of phyllosphere microorganisms. It is taxonomically highly diverse and influences plant physiology, metabolism health. Members the order Diaporthales are distributed worldwide include devastating pathogens as well endophytes saprophytes. However, many species remain uncharacterized, with studies examining their diversity needed. Here, we report on identification several diaporthalean taxa samples collected from diseased leaves Cinnamomum camphora (Lauraceae), Castanopsis fordii (Fagaceae) Schima superba (Theaceae) in Fujian province, China. Based morphological features coupled to multigene phylogenetic analyses internal transcribed spacer (ITS) region, large subunit nuclear ribosomal RNA (LSU), partial beta-tubulin (tub2), histone H3 (his3), DNA-directed polymerase II (rpb2), translation elongation factor 1-α (tef1) calmodulin (cal) genes, three new introduced, namely, Diaporthe wuyishanensis, Gnomoniopsis wuyishanensis Paratubakia schimae. This study contributes our understanding biodiversity fungi that inhabitants trees native Asia.

Abstract This study documents the morphology and phylogeny of micro-fungi collected from various medicinal plants in southern China northern Thailand. Based on morphological characteristics phylogenetic analyses, 100 species are identified distributed three classes (Dothideomycetes, Leotiomycetes Sordariomycetes), 16 orders, 37 families 67 genera. We introduce one new order, Oncopodiellales, families, Catenuliconidiaceae, Diplocladiellaceae Oncopodiellaceae, genus, Biascospora, 54 species: Achroiostachys browniana, A. catenate, fusiformispora, Acrocalymma xishuiense, Amphisphaeria hibiscicola, Apiculospora thailandensis, Arecophila maolanensis, yunanensis, Barriopsis caryotae, Biascospora chishuiensis, Conioscypha synnemata, Diaporthe ervatamiae, D. kunmingensis, tu-chungcola, xishuiensis, hannanensis, Distoseptispora gelatinosa, greeniana, Gregatothecium diflugoscola, Helicosporium multiseptatum, Helminthosporium thailandicum, Kalmusia tetrastigmae, Keissleriella yunnana, Lasiodiplodia houttuyniae, L. poacearum, Leptospora Lophiotrema asexualis, guizhouense, Melanopsamma tongrenensis, Memnoniella chiangmaiensis, Murichromolaenicola dendrobii, Neoheleiosa brownii, Neohelicascus guizhouensis, Neohelicomyces sexualis, Neohendersonia Neomassaria fibraureae, Neoscytalidium Parabahusutrabeeja hyalina, Paramyrothecium Phaeosphaeria boehmeriae, P. guiyangensis, Phaeosphaeriopsis spineleae, Porodiplodia Psiloglonium bambusicola, Roussoella panzhouensis, R. wudangensis, Seriascoma Sphaeropsis Striaticonidium olivaceobrunnea, Tamhinispora obpyriformis, V. chiangraiensis Virgatospora thailandica, with illustrations, discussions their taxonomic placement, comparisons morphologically similar taxa. Pathogen-induced fruit decay is a significant threat to the kiwifruit industry, leading considerable economic losses annually. The cell-wall-degrading enzymes (CWDEs) secreted by these pathogens are crucial for penetrating cell wall and accessing nutrients. Among them, Diaporthe species recognized as major causal agents of soft rot in kiwifruit, yet their pathogenic mechanisms not well understood. In this study, we explored production various CWDEs Z1-1N, including polygalacturonase (PG), polymethylgalacturonase (PMG), polygalacturonic acid transeliminase (PGTE), pectin methyltranseliminase (PMTE), endoglucanase (Cx), β-glucosidase (β-glu), both liquid cultures within infected tissues. Our findings revealed activities two pectinases (PG PMG) cellulases (Cx β-glu) contrast, very low levels PMTE PGTE were observed under same conditions. When orange served carbon source, PG PMG showed notable activities, while remained inactive. Moreover, Cx β-glu significantly decreased more than 63 times medium with carboxymethyl cellulose (CMC) source compared kiwifruit. A further analysis indicated that necrotic lesions produced pectinase extracts larger those cellulase extracts. Notably, four enzymes—PG, PMG, Cx, β-glu—exhibited high on third or fourth day post-infection Z1-1N. These results suggest Z1-1N secretes range contribute enhancing PG, β-glu. This study sheds light pathogenicity highlights importance process.

Endophytic fungi associated with four plant species in Korea were isolated and characterized using morphological molecular analyses. Phylogenetic analyses of the internal transcribed spacer (ITS) region, β-tubulin (TUB) gene, translation elongation factor 1-α (EF1-α) gene revealed previously unrecorded Diaporthe Korea: D. caryae, phoenicicola, stewartii, unshiuensis. Species determination in the Amidella clade is notoriously difficult, because of relative dearth diagnostic characters and rather common occurrence homoplasies. This results a substantial number misnamed unnamed collections, misapprehension geographic range known species, gross underestimation species it contains. To assess diversity that should be considered as part Amidella, DNA sequences available for this group were retrieved public nucleotide databases, using combination approaches to achieve comprehensive representation. Phylogenetic analysis based on aligned ITS sequences, consistently with from other molecular markers (ncLSU, RPB2, TEF1, BTUB), suggests five major clades: one containing type Amanita volvata, another ponderosa allies, third (roughly half all species) lepiotoides, two others without valid yet. At level, around 81 clades delimited, which only 16-17 can assigned name, few more corresponding provisional taxa listed amanitaceae.org website. Up three further might correspond proposed clades. The current evidence narrow most these study provides phylogeny-arranged outlook worldwide distribution an infrasectional framework optimizing taxonomic sampling designing clade-specific assist identification.
