Background: Disulfiram (DSF), an FDA-approved pharmaceutical for the management of alcoholism, has demonstrated its efficacy against several kinds cancer. DSF limited solubility, a fast metabolism, short duration action, and instability in physiological environments, mostly caused by rapid degradation acidic gastric environment. Objective: A transdermal gel containing disulfiram, which was loaded into invasomes, developed to improve stability enable effective distribution tumor tissues. Methods: This study included 72 Wistar rats weighing 200±35 g, were separated two groups, each 12 animals. Rats orally provided dose 5 mg pure suspension via oral gavage, nano-invasomal then applied their skin. is determined rats' plasma reverse-phase high-performance liquid chromatography (RP-HPLC). Results: The results showed that maximum effect (Cmax, Tmax, AUC0-72) (Cmax=57.3±0.2, Tmax=3.6±0.01 562±3. 3ng.h/ml) (Cmax=138±0.4, Tmax=5.5±0.01 2819±6.6 ng. h/ml) routes, respectively. Results time concentration needed achieve (Cmax Tmax) significantly different between DSF-oral invasomal (p<0.05). relative bioavailability route five times after single administered hours. Conclusions: filled with more convenient way administering compared route.
