New approaches to the treatment of women with ovarian cancer are desperately needed, since most develop resistance chemotherapy and 5-year survival rate remains low. The hypothesis guiding this study was that inhibition cell adhesion could be used as a novel strategy increase chemosensitivity cells. Nectin-4 peptide N4-P10 inhibit formation cell-cell aggregates (spheroids) using lines cells isolated from patients' ascites. Cell were pre-treated or control scrambled peptides monitored for spheroid live-cell imaging by digital time-lapse photography. Cells then tested cytotoxicity chemotherapeutic agent, cisplatin. Peptide blocked aggregation in different levels expression morphologies. cisplatin increased N4-P10. Similarly, when single ascites patients, These results suggest targeting adhesive property serve new approach augment cytotoxic effect potentially reduce disease recurrence patients.
