Abstract Background Diabetic retinopathy (DR) is the foremost cause of vision loss among global working-age population, and statins are most frequently prescribed drugs for lipid management in patients with DR. The exact relationship between DR has not been determined. This study sought to validate causal association usage diabetic retinopathy. Methods summary-data-based Mendelian randomization (SMR) method inverse-variance-weighted (IVW-MR) were used identify via use expression quantitative trait loci (eQTL) data 3-hydroxy-3-methylglutaryl-coenzyme A reductase (HMGCR) (31,684 blood samples), low density lipoprotein cholesterol-related GWAS (sample size: 440,546), DR-related (14,584 cases 176,010 controls). Additionally, a cross-sectional observational based on from National Health Nutrition Examination Survey (NHANES) was conducted supplement 106,911). odds ratios (ORs) corresponding 95% confidence intervals (CIs) employed evaluate results. Results Based results MR analysis, HMGCR inhibitors causally connected noticeably greater incidence (IVW: OR = 0.54, CI [0.42, 0.69], p 0.000002; SMR: 0.66, [0.52, 0.84], 0.00073). Subgroup analysis revealed that affected by severity sensitivity stability reliability NHANES also support taking decreased risk (OR [0.37, 0.79], 0.001). Conclusions significant increase related use, providing novel insights into role However, further investigations needed verify these findings.

Abstract Background As the population ages, mild cognitive impairment (MCI) and type 2 diabetes mellitus (T2DM) become common conditions that often coexist. Evidence has shown MCI could lead to reduced treatment compliance, medication management, self‐care ability in T2DM patients. Therefore, early identification of those with increased risk is crucial from a preventive perspective. Given growing utilization decision trees prediction health‐related outcomes, this study aimed identify patients using tree approach. Methods This hospital‐based case–control was performed Endocrinology Department Xiangya Hospital affiliated Central South University between March 2021 December 2022. defined based on Petersen criteria. Demographic characteristics, lifestyle factors, T2DM‐related information were collected. The sample randomly divided into training validation sets 7:3 ratio. To explore the causal associations among circulating proteins, plasma metabolites, and age-related macular degeneration (AMD). We employed Mendelian randomization (MR) analysis colocalization to discern relationship between proteomes AMD. This investigation utilized data from protein quantitative trait loci (pQTL) studies in deCODE UK Biobank. Additionally, metabolite-related genome-wide association (GWAS) AMD-related GWAS were incorporated. Our findings confirmed a potential cytoplasmic tryptophanyl-tRNA synthetase 1 (WARS1) higher risk of The observed impact WARS1 on two subtypes AMD (dry wet) align consistently with aforementioned outcomes. Three metabolites-N-acetyl-kynurenine, N-acetyltyrosine, caproate (6:0)-were identified as mediators effect AMD, subgroup revealed that N-acetyltyrosine is specific negative metabolite associated dry whereas X-16580 positive linked wet outcomes this study suggest role proteins mediating metabolites by integrating multiple genetic analyses. Nevertheless, further research essential validate strengthen these conclusions. establishes

Background Pancreatitis is a serious and complex inflammatory disease that imposes severe effect on quality of life. Links between plasma lipidome pancreatitis have been reported, some which not yet clearly elucidated. Methods Therefore, our study aimed to investigate the causal relationships four types by conducting bidirectional, two-sample Mendelian randomization (MR) analysis. We obtained genetic variants associated with 179 lipid species from Genome-wide association analysis lipidome. The aggregated statistical data acute (AP), alcohol-induced (AAP), chronic (CP), (ACP) FinnGen consortium were exploited as outcome. inverse variance weighted (IVW) technique main method was used for MR sensitivity analyses evaluate heterogeneity pleiotropy. The association between myopia and diabetic retinopathy (DR) is unclear, with inconsistent results reported, whether the represents causality remains unknown. This study aimed to investigate causal associations of genetically determined DR, further explore specific mechanisms. We conducted two-sample mendelian randomization (MR) analyses any high on six DR phenotypes, including background severe proliferative (PDR), maculopathy unspecific in primary study. Mechanism exploration spherical equivalent refraction (SER), corneal curvature (CC) axial length (AL) was carried out subsequently. Single-nucleotide polymorphisms (SNPs), used as genetic instruments, were derived from UK Biobank, Genetic Epidemiology Research Adult Health Aging cohort (GERA) FinnGen. inverse variance weighted (IVW) method mainly assess causality, complemented sensitivity direction analyses. Using SNPs that have excluded possible confounders, we discovered suggestive positive (IVW: odds ratio [OR] = 1.133, 95% confidence interval [95%CI]: 1.070-1.201, P 1.91×10-5) PDR OR 1.182, CI: 1.088-1.285, 8.31×10-5). Similar but more significant found 1.107, 95%CI: 1.051-1.166, 1.39×10-4; 1.163, 1.088-1.244, 8.76×10-6, respectively). Further mechanism only AL, rather than SER or CC, strongly significantly associated DR. These robust risk PDR, which might be related indicating significance control for preventing development progression.
