The 6th Ottawa International Conference in Neuromuscular Disease and Biology was held on September 7–9, 2023 Ottawa, Canada. goal of the conference to assemble international experts fundamental science, translational medicine clinical neuromuscular disease research. Speakers provided attendees with updates a wide range topics related biology, including methods identify novel diseases, recent developments muscle, motor neuron stem cell expanded pathogenesis known exciting advances therapy development. Duchenne muscular dystrophy (DMD) is a lethal disease caused by mutations in the DMD gene that encodes dystrophin. Dystrophin deficiency also impacts muscle stem cells (MuSCs), resulting impaired asymmetric cell division and myogenic commitment. Using MuSCs from patients mouse model mdx , we found PTPN1 phosphatase expression up-regulated STAT3 phosphorylation concomitantly down-regulated MuSCs. To restore STAT3-mediated signaling, examined effect of K884, novel PTPN1/2 inhibitor, on Treatment with K884 enhanced promoted differentiation patient-derived In mice, treatment increased number divisions, correlating differentiation. Interestingly, pro-myogenic specific to human murine absent control Moreover, loss-of-function experiments indicate impact mediated mainly through PTPN1. We propose inhibition may serve as therapeutic strategy function DMD.
