Abstract This study aimed to investigate the potential hypoglycemic mechanism of red ginseng acidic polysaccharides (RGAP) from perspective fatty acid (FA) regulation. A high‐glucose/high‐fat diet in conjunction with streptozotocin administration was employed establish type 2 diabetes mellitus (T2DM) rat models, and their fecal FAs were detected using liquid chromatography‐mass spectrometry (LC‐MS) method. RGAP treatment alleviated polyphagia, polydipsia, weight loss, hyperglycemia observed T2DM rats. FA profile disturbed by modeling, 11 marker selected statistical analysis, whose intensities reversely changed administration. Among these FAs, short‐chain negatively correlated fasting blood glucose (FBG) level, while positive correlations between long‐chain FBG level. Combined metabolite‐enzyme‐gene network we inferred that mechanistic on may be associated regulation metabolism inflammation‐related signaling pathways. confirmed regulatory effect FA, which can provide a scientific basis new ideas for developing as functional food supplementary T2DM.

ABSTRACT Liquid chromatography‐mass spectrometry (LC‐MS) has become an indispensable tool for elucidating molecular structures and quantifying diverse compounds within complex mixtures. Despite its versatility, it faces various challenges such as ion suppression, low sensitivity, analyte instability, matrix effects, which are being overcome by different kinds of offline online derivatization techniques to improve specificity reduce potential interferences. In this context, considerable advancements have been made in reviewing critically evaluating a wide range developed methods techniques; however, little attention given post‐column (PCD) LC‐MS. Therefore, comprehensive review highlights state‐of‐the‐art LC‐MS with specific focus on types chemical physical PCD, in‐source derivatization. Many biologically active metabolites of the essential amino acid L-tryptophan (Trp) are associated with different neurodegenerative diseases and neurological disorders. Precise reliable methods for their determination needed. Variability in physicochemical properties makes analytical process challenging. In this case, chemical modification analyte derivatization could come into play. Here, we introduce a novel fast reversed-phase ultra-high-performance liquid chromatography (RP-UHPLC) coupled tandem mass spectrometry (MS/MS) method Trp its ten human plasma samples after 2-bromo-4′-nitroacetophenone (BNAP). The procedure was optimized terms incubation time, temperature, concentration, volume reagent. Method development comprises choice suitable stationary phase, mobile phase composition, gradient elution optimization. developed validated according to ICH guidelines. Results all validation parameters were within acceptance criteria guideline, i.e., intra- inter-day precision (expressed as relative standard deviation; RSD) range 0.5–8.2% 2.3–7.4%, accuracy 93.3–109.7% 94.7–110.1%, limits detection (LODs) 0.15–9.43 ng/mL, coefficients (R2) higher than 0.9906, carryovers were, cases, less 8.8%. practicability evaluated using blue applicability grade index (BAGI) score 65.
