Abstract Fumarole fields on active volcanoes are habitats that host unique microbial ecosystems. However, DNA extraction from them for further analysis is rather challenging. In this study, we compared two different ways of sample homogenization to profile the communities fumarolic Elbrus and Ushkovsky frozen fumarole deposits Fujiyama. Vertical homogeniser gave significantly higher concentrations samples, more archaeal amplicon sequence variants (ASVs) samples horizontal one. This suggests vertical might be preferable sandy rocky soils. Independent homogenizer type, dominant phyla were Acidobacteriota Pseudomonadota, Crenarchaeota Ushkovsky. The bacterial community Fuji was less diverse, with Actinomycetota, Pseudomonadota Bacillota being phyla. Thus, studied fumaroles showed distinct profiles, revealing adaptations their respective extreme environments. Within fungal community, Ascomycota, Basidiomycota Chytridiomycota most all three volcanoes, but abundance varied. study offers first comprehensive fields, demonstrates choice methodology can influence understanding diversity in
