The intestinal barrier maintained by various types of columnar epithelial cells, plays a crucial role in regulating the interactions between contents (such as microbiota), immune system, and other components. Dysfunction mucosa is significant pathophysiological mechanism clinical manifestation inflammatory bowel disease (IBD). However, current therapies for IBD primarily focus on suppressing inflammation, no disease-modifying treatments specifically target barrier. Given side effects associated with chronic immunotherapy, effective alternative that promote mucosal healing are highly attractive. In this review, we examined function mechanisms behind its disruption IBD. We illustrated complex process proposed therapeutic approaches to strategies These included application stem cell transplantation organ-like tissue engineering generate new tissue. Finally, discussed potential restore treatment

Lung inflammation is an essential inducer of various diseases and closely related to pulmonary-endothelium dysfunction. Herein, we propose a pulmonary endothelium-targeted codelivery system anti-inflammatory indomethacin (IND) antioxidant superoxide dismutase (SOD) by assembling the biopharmaceutical SOD onto “vector” rod-like pure IND crystals, followed coating with anti-ICAM-1 antibody (Ab) for targeting endothelial cells. The has 237 nm diameter in length extremely high drug loading 39% 2.3% SOD. Pharmacokinetics biodistribution studies demonstrate extended blood circulation strong accumulation after intravenous injection lipopolysaccharide (LPS)-induced inflammatory murine model. Particularly, allows robust capacity target endothelium mostly due rod-shape Ab effect. In vitro, preparation shows synergistic effects LPS-activated vivo, exhibits superior pharmacodynamic efficacy revealed significantly downregulating inflammatory/oxidative stress markers, such as TNF-α, IL-6, COX-2, reactive oxygen species (ROS), lungs. conclusion, based on crystals could well effectively alleviate lung inflammation. study offers promising approach combat endothelium-associated diseases.
