Despite the tremendous clinical potential of nucleic acid–based vaccines, their efficacy to induce therapeutic immune response has been limited by lack efficient local gene delivery techniques in human body. In this study, we develop a hydrogel-based organic electronic device (μEPO) for both transdermal acids and vivo microarrayed cell electroporation, which is specifically oriented toward one-step transfection DNAs subcutaneous antigen-presenting cells (APCs) cancer immunotherapy. The μEPO contains an array microneedle-shaped electrodes with pre-encapsulated dry DNAs. Upon pressurized contact skin tissue, are rehydrated, electrically triggered release DNAs, then electroporate nearby cells, can achieve more than 50% epidermal upper dermal layer. The emergence of immunotherapy as a revolutionary therapeutic modality has fostered confidence and underscored its potent efficacy in tumor therapy. However, enhancing the by precise judicious administration poses significant challenge. In this context, we have developed disulfide-bearing transdermal nanovaccine integrating thiol-reactive agent lipoic acid (LA) into metal-coordinated cyclic dinucleotide nanoassembly, designated LA-Mn-cGAMP (LMC) nanovaccines. Upon topical application to skin with melanoma, dithiolane moiety LA enables thiol-disulfide dynamic exchange skin, hence facilitating penetration both subcutaneous tissues via thiol-mediated uptake (TMU) mechanism. Our findings demonstrate that LMC significantly enhances STING activation, mitigates immunosuppressive microenvironment (TME), retards melanoma progression. Moreover, remodeled TME amplifies immune checkpoint inhibitors. This advancement offers an strategy for existing agonist therapy, potentially improving biosafety immunotherapy.
