Garlic polysaccharide (GP) is an inulin-type fructan with potent antioxidant activity, whereas its health-promoting functions are not well explored. In the current study, we established a fatigue model by forcing mice to swim in water tank for at least one hour every day. We measured changes body weight, exhaustive swimming time, and biochemical indices related weight-bearing fed low (1.25 g/kg-BW) high doses (2.5 of GP daily gavage 7 weeks. The soluble was extracted from industrial garlic wastes using patented method. results indicate that improved symptoms increasing duration swimming, restoring blood markers (BUN BLA), liver muscle glycogen reserves fatigued mice. also increased enzyme activity (SOD, GSH-Px, CAT) restored ATPase activating AMPK/PGC-1α pathway. Additionally, modified gut microbiota potentially beneficial bacteria decreasing harmful bacteria. increase Bacteroidota decrease Firmicutes phyla regulate metabolism short-chain fatty acids gut. conclusion, may be effective alleviating exercise-induced through multiple mechanisms can developed into health anti-fatigue supplements.

Hyperglycemia and hyperlipidemia are the hallmarks of type 2 diabetes mellitus (T2DM). T2DM is a systemic metabolic disease caused by insulin resistance malfunctioning pancreatic β-cells. Although ginseng (the roots Panax C.A. Meyer) can be used to treat T2DM, underlying mechanism unclear. To assess role of, γ-aminobutyric acid-fructosyl-glucose (GABAFG), maillard reaction product ginseng, in treatment. The metabolism GABAFG serum tissues was analyzed via ultra-high performance liquid chromatography-Q exactive-mass spectrometry (UHPLC-QE-MS). molecular mechanisms β-cells (in vivo vitro) were investigated Western blotting, qPCR immunofluorescence. In addition, results validated high-throughput sequencing metabolomics. alleviated elevation blood glucose lipids HFD/STZ-induced mice. Also, reduced resistance-associated IRS-1 signaling axis vitro. Mechanistically, targeted nuclear translocation TFEB inhibited apoptosis enhancing autophagolysosome function. remodeled gut microbiota. Specifically, increased Akkermansia, decreased Romboutsia abundance, glycerophospholipid metabolism, thus alleviating T2DM-induced dyslipidemia. This first study pharmacological effects ginseng-derived T2DM. Therefore, this provides new theoretical basis for understanding effect diseases.

Cancer immunotherapy has revolutionized cancer treatment, but therapeutic ineffectiveness-driven by the tumor microenvironment and immune evasion mechanisms-continues to limit its clinical efficacy. This challenge underscores need explore innovative approaches, such as multimodal immunotherapy. Phytochemicals, bioactive compounds derived from plants, have emerged promising candidates for overcoming these barriers due their immunomodulatory antitumor properties. review explores synergistic potential of phytochemicals in enhancing modulating responses, reprogramming microenvironment, reducing immunosuppressive factors. Integrating with conventional strategies represents a novel approach mitigating resistance outcomes. For instance, nab-paclitaxel shown checkpoint inhibitors, while QS-21 synergistically enhances efficacy vaccines. Furthermore, we highlight recent advancements leveraging nanotechnology engineer improved bioavailability targeted delivery. These innovations hold great promise optimizing application phytochemicals. However, further large-scale studies are crucial fully integrate into immunotherapeutic regimens effectively.

ABSTRACT Ulcerative colitis (UC) is a recurrent, chronic disease whose main symptoms include weight loss, diarrhea, and blood in the stool. In recent years, incidence of UC has been increasing year by year, which seriously affects daily life patients. Luteolin (Lut), as flavonoid, widely found variety vegetables fruits shown to have pharmacological activities. This work investigated effects Lut on dextrose sodium sulfate (DSS)‐induced ulcerative mice, with special focus role endoplasmic reticulum (ER) stress this. The outcomes demonstrated that symptoms, including phenotype, elevated inflammatory factor levels, intestinal barrier damage, gut microbiota disruption, were considerably alleviated model mice treated luteolin. Also, ER apoptosis mice. We then explored induced thapsigargin (TG) tunicamycin (TM) HT29 cells vitro. It was treatment inhibited TM/TG‐induced apoptosis. However, these inhibitory attenuated SIRT1 silencing TM‐treated cells. conclusion, our results suggest supplementation mouse improves provides theoretical basis for further application prevention inflammation‐related diseases humans.

The aryl hydrocarbon receptor (AhR) plays a crucial role in regulating many physiological processes. Activating the AhR-CYP1A1 axis has emerged as novel therapeutic strategy against various inflammatory diseases. Here, practical situ cell-based fluorometric assay was constructed to screen modulators, via functional sensing of CYP1A1 activities live cells. Firstly, cell-permeable, isoform-specific enzyme-activable fluorogenic substrate for rationally in-situ visualizing dynamic changes function living systems, which subsequently used discovering efficacious modulators axis. Following screening compound library, LAC-7 identified an activator axis, dose-dependently up-regulated expression levels both and AhR multiple cell lines. also suppressed macrophage M1 polarization reduced factors LPS-induced bone marrow-derived macrophages. Animal tests showed that could significantly mitigate DSS-induced ulcerative colitis acute lung injury mice, markedly factors. Collectively, optimized devised imaging strongly facilitated discovery anti-inflammatory agents.
