The zig-zag eel (Mastacembelus armatus) is a unique economic fish species in China and exhibits significant dimorphism of male female phenotypes. Cultivating all-male seedlings can significantly improve production efficiency. To investigate sex differentiation gonadal development M. armatus, high-throughput sequencing technology was used to analyze the transcriptomes gonads at different developmental stages, both before after differentiation. We identified key genes involved differentiation, male-specific differentially expressed (DEGs), including dmrt1, amh, sox9, gsdf, dmrt2b, female-biased DEGs, foxl2, rspo1, gdf9, bmp15, wnt4. GO KEGG enrichment analyses revealed that signaling pathways such as MAPK, Wnt, TGF-β play roles armatus. expression levels 13 sex-related genes, wnt4, were determined by RT–qPCR addition RNA sequencing. RT-qPCR validation results consistent with transcriptomic data, confirming reliability our findings. This research provides valuable insights into mechanisms armatus lays foundation for developing populations aquaculture.

Female common carp grow faster than male individuals, implying that rearing females could be more profitable in aquaculture. Non-coding RNAs (ncRNAs) serve as versatile regulators with multiple functions diverse biological processes. However, the roles of ncRNAs sex differentiation are less studied. In this study, we investigated expression profiles ncRNAs, including miRNAs, lncRNAs, and circRNAs, gonads to comprehend carp. A substantial number differentially expressed (DE) ovaries testes were identified. Some notably miR-205, miR-214, miR-460-5p, might modulate hormone synthesis thus maintain sex. novel miRNA, novel_158, was predicted suppress foxl3. DE lncRNAs associated oocyte meiosis, GnRH signaling pathways, steroid biosynthesis, while circRNA target genes enriched WNT pathway MAPK pathway. We also analyzed ncRNA-mRNA interactions shed light on crosstalk between competing endogenous (ceRNAs), which is critical mechanism by circRNAs function. may able competitively bind novel_313, a new regulate hsd17β3. Our research will provide valuable resource for understanding genetic basis gonadal development

Epigenetic modifications are critical in precisely regulating gene expression. The common carp (Cyprinus carpio) is an economically important fish species, and females exhibit faster growth rates than males. However, the studies related to epigenetic gonads limited. In this study, we conducted Assay for Transposase Accessible Chromatin sequencing (ATAC-seq) Bisulfite (BS-seq) explore roles of gonads. We identified 84,207 more accessible regions 77,922 less ovaries compared testes, some sex-biased genes showed differential chromatin accessibility their promoter regions, such as sox9a zp3. Motif enrichment analysis that transcription factors (TFs) associated with embryonic development cell proliferation were heavily enriched ovaries, TFs Foxl2 SF1 only ovaries. also analyzed possible regulations between By BS-seq, 2087 differentially methylated (promoter-DMGs) 5264 body (genebody-DMGs) CG contexts. These genebody-DMGs significantly Wnt signaling pathway, TGF-beta GnRH indicating methylation could play essential role sex maintenance, just like regions. Combined transcriptomes, revealed expression dmrtb1-like, spag6, fels was negatively correlated levels Our study on contributes elucidating molecular mechanism differentiation maintenance carp.
