Ovarian aging typically precedes the decline of other organ systems, yet its molecular mechanisms remain poorly understood. Glycosylation as one most important protein modifications has been especially unexplored in this context. Here, we present first high-resolution glycoproteomic landscape mouse ovaries, uncovering site-specific N-glycan signatures across subcellular components such high proportions complex glycans, core fucosylation, and LacdiNAc branches at zone pellucida. We report three major glycosylation alterations aged ovaries: frequently changed core-fucosylation associated with cell adhesion immune responses, decreased glycans on zona pellucida (ZP) responsible for fertility decline, increased sialylated modified by Neu5Ac Neu5Gc playing different roles activation responses. Integrated multi-omic analyses further highlight unique role glycosylation, distinct from phosphorylation, regulating key signaling pathways, antigen processing presentation, complement coagulation cascades, ROS biosynthetic metabolic processes, well death. This study offers a novel glycobiological perspective ovarian aging, broadening our understanding beyond traditional approaches.

Aging is a complex, progressive, and irreversible biological process that entails numerous structural functional changes in the organism. These affect all bodily systems, reducing their ability to respond adapt environment. Chronic inflammation one of key factors driving development age-related diseases, ultimately causing substantial decline abilities older individuals. This persistent inflammatory state (commonly known as “inflammaging”) characterized by elevated levels pro-inflammatory cytokines, an increase oxidative stress, perturbation immune homeostasis. Several factors, including cellular senescence, contribute this milieu, thereby amplifying conditions such cardiovascular disease, neurodegeneration, metabolic disorders. Exploring mechanisms chronic aging essential for developing targeted interventions aimed at promoting healthy aging. review explains strong connection between inflammation, highlighting potential therapeutic approaches like pharmacological treatments, dietary strategies, lifestyle changes.

The hypothalamus is a critical organ that regulates sexual development in animals. However, current research on the hypothalamic regulation of maturation female goats remains limited. In this study, we conducted metabolomic and transcriptomic analyses tissues Jining grey at different stages (1 day old (neonatal, D1, n = 5), 2 months (prepuberty, M2, 4 (sexual maturity, M4, 6 (breeding period, M6, 5)). A total 418 differential metabolites (DAMs) were identified among which abundance such as anserine, L-histidine, carnosine, taurine, 4-aminobutyric gradually increased with progression development. These may regulate neuronal hormone secretion processes by influencing metabolism histidine phenylalanine. Through combined analyses, differentially expressed genes mitogen-activated protein kinase 9 (MAP3K9), prune homolog BCH domain (PRUNE2), potassium voltage-gated channel interacting 4(KCNIP4) jointly energy Gonadotropin-releasing neurons conjunction DAMs, including LPC22:5, 2-Arachidonyl Glycerol ether, LPE22:5, Lysops22:5. Additionally, elucidated molecular mechanism through glutathione goats. summary, study illustrates dynamic changes mRNA within tissue during postnatal This provide significant scientific insights for future animal breeding.

Background Evidence suggests a connection between DNA methylation (DNAm) aging and reproductive aging. However, the causal relationship DNAm age at menopause remains uncertain. Methods Employing established epigenetic clocks, such as Hannum acceleration (Hannum), Intrinsic (IEAA), DNAm-estimated granulocyte proportions (Gran), GrimAge (GrimAgeAccel), PhenoAge (PhenoAgeAccel), plasminogen activator inhibitor-1 levels (DNAmPAIadjAge), bidirectional Mendelian randomization (MR) study was carried out to explore potential causality menopausal age. The primary analytical method used inverse variance weighted (IVW) estimation model, supplemented by various other techniques. Results or deceleration, indicated Hannum, IEAA, Gran, GrimAgeAccel, PhenoAgeAccel, DNAmPAIadjAge, did not exhibit statistically significant effect on according forward MR analysis. there suggestive positive association Gran (Beta = 0.0010; 95% confidence interval (CI): 0.0004, 0.0020) in reverse Conclusion observed increase relation could potentially serve valuable indicator for evaluating physiological status onset of menopause.
