Iron has been shown to trigger oxidative stress by elevating reactive oxygen species (ROS) and participate in different modes of cell death, such as ferroptosis, apoptosis necroptosis. However, whether iron-elevated ROS is also linked pyroptosis not reported. Here, we demonstrate that iron-activated can induce via a Tom20-Bax-caspase-GSDME pathway. In melanoma cells, iron enhanced signaling initiated CCCP, causing the oxidation oligomerization mitochondrial outer membrane protein Tom20. Bax recruited mitochondria oxidized Tom20, which facilitates cytochrome c release cytosol activate caspase-3, eventually triggering pyroptotic death inducing GSDME cleavage. Therefore, acts causative factor Tom20 senses for iron-driven cells. Since activates GSDME-dependent induction cells specifically express high level GSDME, may be potential candidate therapy. Based on functional mechanism above, further supplementation at dosage used iron-deficient patients sufficient maximize anti-tumor effect clinical ROS-inducing drugs inhibit xenograft tumor growth metastasis through pyroptosis. Moreover, no obvious side effects are observed normal tissues organs mice during combined treatment iron. This study only identifies sensitizer amplifying drive pyroptosis, but implicates novel iron-based intervention strategy

Heavy metals bring long-term hazardous consequences and pose a serious threat to all life forms. Being non-biodegradable, they can remain in the food webs for long period of time. Metal ions are essential indispensable almost aspects metabolism but be toxic beyond threshold level living beings including microbes. generally present environment, many geogenic anthropogenic activities has led excess metal ion accumulation environment. To survive harsh contaminated environments, bacteria have certain resistance mechanisms metabolize transform heavy into less This also gives rise different species resistant bacteria. Ferroptosis is a novel type of programmed cell death, characterized by dysregulated iron metabolism and accumulation lipid peroxides. It features the alteration mitochondria aberrant excessive as well loss cysteine-glutathione-GPX4 axis. Eventually, accumulated peroxides result in lethal damage to cells. induced overloading ROS can be inhibited activation GPX4 pathway, FS1-CoQ10 GCH1-BH4 DHODH it also regulated oncogenes tumor suppressors. involves various physiological pathological processes, increasing evidence indicates that ferroptosis play critical role cancers other diseases. inhibits proliferation malignant cells types inducing may become new method cancer treatment. Many inhibitors targeting key factors such SLC7A11, GPX4, overload have been developed. The application mainly divided into two directions, i.e. avoid healthy selectively induce cancers. This review delves into the intricate relationship between anemia, iron metabolism, and human immunodeficiency virus (HIV), aiming to unravel interconnected pathways that contribute complex interplay these 3 entities. A systematic exploration of relevant literature was conducted, encompassing studies examining association status, HIV infection. Both clinical preclinical investigations were analyzed elucidate underlying mechanisms linking components. Chronic inflammation, a hallmark infection, disrupts homeostasis, impacting erythropoiesis contributing anemia. Direct viral effects on bone marrow function further compound red blood cell deficiencies. Antiretroviral therapy, while essential for managing HIV, introduces potential complications, including medication-induced Dysregulation levels in different tissues adds complexity network interactions. Effective management anemia necessitates multifaceted approach. Optimization antiretroviral treatment opportunistic infections, targeted nutritional interventions, supplementation, are integral However, challenges persist understanding specific molecular governing pathways. Decoding is imperative enhancing holistic care individuals with HIV/AIDS. nuanced relationships will inform development more precise optimizing this population. Future research endeavors should focus elucidating mechanisms, paving way innovative therapeutic strategies context HIV-associated
