Background Zingiber officinale Roscoe has been shown to possess analgesic properties. Zingerone (ZO), a bioactive compound derived from Roscoe, exhibits range of pharmacological effects, including anti-inflammatory, anti-cancer, antioxidant, antibacterial, and anti-apoptotic activities. However, the properties zingerone remain unclear. Methods Complete Freund’s adjuvant (CFA) was administered left hind paw C57BL/6 mice induce model inflammatory pain. The effects were assessed using Von Frey Hargreaves tests. In vivo fiber photometry whole-cell patch clamp techniques employed investigate potential mechanisms. Results Both acute long-term treatment with resulted in significant increase mechanical thermal pain thresholds experiencing CFA-induced Mechanical stimulation led pronounced calcium levels within anterior cingulate cortex (ACC) neurons model, which alleviated by administration. Furthermore, found modify synaptic transmission ACC decrease their intrinsic excitability prolonging refractory period these neurons. Conclusion demonstrates for alleviating reducing mouse model.
