Gallbladder cancer (GBC) is a highly aggressive malignancy exhibiting correlation between increased body mass index and risk of developing GBC. In obese individuals, the release free fatty acids from adipose tissue into circulating blood augmented. However, role oleic acid (OA), one most abundant monounsaturated in plasma, GBC cell proliferation has not been determined. this study, we investigated effects OA on cells. We focused G protein-coupled receptor 120/free 4 (GPR120/FFAR4) 40/free 1 (GPR40/FFAR1), which have high affinity for long-chain acids. significantly promoted human lines (G-415 GBC-SD) vitro, with highest increase observed at 200 μM OA. vivo, OA-treated nude mice bearing G-415 xenografts exhibited enhanced tumor growth compared to controls. Immunohistochemical analysis revealed expression GPR120 GPR40 cultured cells patient tissues. OA-induced was mediated by GPR120, as evident reduced upon silencing or inhibition, no effect inhibition. Furthermore, activation ERK phosphorylation, implicating GPR120/ERK signaling pathway growth. To our knowledge, first study elucidate via suggesting its potential therapeutic target treatment.
