Major
depressive
disorder
(MDD)
is
one
of
the
most
prevalent
psychiatric
disorders,
with
an
increasing
incidence
each
year
and
important
socioeconomic
burden.
Although
new
treatments
are
continuously
being
developed,
there
no
effective
monitoring
method
to
determine
suitability
treatment
ensure
positive
outcomes.
Therefore,
patients
often
struggle
ineffective
antidepressants
their
potential
adverse
effects,
which
halts
any
future
progress
in
managing
disorder.
Considering
microRNAs
(miRNAs)
as
biomarkers
for
various
pathologies

Abstract
Apolipoprotein
E
(apoE)
genotyping
is
a
valuable
tool
for
assessing
genetic
risk
associated
with
Alzheimer’s
disease
(AD).
The
ε4
allele
strongly
linked
to
an
increased
and
earlier
onset
of
AD
symptoms.
Individuals
carrying
one
(heterozygous)
or
two
(homozygous)
copies
the
are
at
significantly
higher
compared
those
without
it.
In
contrast,
ε2
may
offer
protective
effect,
reducing
likelihood
developing
disease.
Many
studies
have
reported
use
PCR-based
methods
apoE
genotyping.
However,
current
approaches
typically
require
least
tubes—and
often
more—to
distinguish
among
six
possible
genotypes
using
mixtures
allele-specific
primers.
Consolidating
assay
into
single
tube
can
substantially
reduce
time,
minimize
manual
labor,
increase
throughput.
To
address
this
need,
we
developed
one-pot
PCR
platform
genotyping,
utilizing
RNA-DNA
hybrid
CataCleave
probe.
We
provide
detailed
explanation
assay’s
principles,
including
its
limit
detection,
specificity,
resistance
interference
from
other
serum
components,
precision.
Collectively,
information
enables
researchers
adopt
our
streamlined,
single-tube
method
ease.
