Abstract Neutrophils, the most abundant type of granulocyte, are widely recognized as one pivotal contributors to acute inflammatory response. Initially, neutrophils were considered mobile infantry innate immune system, tasked with immediate response invading pathogens. However, recent studies have demonstrated that versatile cells, capable regulating various biological processes and impacting both human health disease. Cytokines other active mediators regulate functional activity by activating multiple receptors on these thereby initiating downstream signal transduction pathways. Dysfunctions in disruptions neutrophil homeostasis been implicated pathogenesis numerous diseases, including cancer disorders, often due aberrant intracellular signaling. Gastrointestinal (GI) cancers, which mainly include malignancies of the esophagus, stomach, intestine, pancreas, liver, gallbladder, and bile duct, pose a significant global health burden. Unfortunately, prognosis for most GI cancers remains poor, particularly in advanced stages. Current treatment options, including targeted immunotherapies, are less effective compared to those other cancer types, highlighting an urgent need novel molecular targets. NEK (NIMA related kinase) kinases group serine/threonine (NEK1-NEK11) that play role regulating cell cycle, mitosis, various physiological processes. Recent studies suggest several members overexpressed human gastrointestinal can contribute tumor progression drug resistance. Among these, NEK2 stands out its consistent overexpression all types cancer. Targeting with specific inhibitors has shown promising results preclinical studies, gastric pancreatic cancers. The development clinical evaluation have emerged as therapeutic strategy. Specifically, inhibitor, T-1101 tosylate, is currently undergoing trials. Inflammatory pathways are often hijacked by cancer cells to favor their own proliferation and survival. Cysteine dioxygenase type 1 (CDO1), an iron-dependent thiol enzyme, catalyzes the rate-limiting step for cysteine oxidation, so that functions as important regulator of cellular availability. However, whether inflammatory environment affects CDO1 activity oxidation its potential impact on tumor growth remains substantially elusive. In present study, we demonstrate is inhibited upon IL-6 treatment, without noticeable alterations in expression. Mechanistically, AKT1 phosphorylates T89 under which represses enzymatic disrupting iron incorporation. Further, AKT1-mediated phosphorylation required IL-6-elicited oral squamous cell carcinoma (OSCC) growth, associated with progression OSCC development.
