Glycoproteins are of significant value to liquid biopsy human diseases. Herein, we present a universal electrochemical platform for the amplified detection glycoproteins, taking advantage glycan-matchmade multivalent decoration enzyme labels enzymatic signal amplification. Briefly, involves two steps, i.e., site-directed phenylboronic acid-coated gold nanoparticles (PBA-AuNPs) cis-diol-containing glycans glycoproteins and subsequent via affinity cross-linking between glycan moieties remaining PBA groups on PBA-AuNP cross-linkers. As matchmaking-based strategy enables each glycoprotein with multiple labels, this exhibits high sensitivity toward detection. Using alkaline phosphatase (ALP) as proof-of-concept label in combination solid-state voltammetric stripping assay enzymatically deposited metallic silver, limits at pg mL-1 level have been obtained aptamer-based thrombin prostate-specific antigen. Overall, work illustrates an efficient versatile ultralow concentration levels, holding desirable advantages simplicity cost-effectiveness over sandwich enzyme-linked immunosorbent assays.
