Emphasizing the significance of cancer-associated fibroblasts (CAFs), non-malignant yet pivotal players within tumor microenvironment (TME), this review illuminates role inflammatory subtype (iCAF) as catalysts in cancer proliferation, metastasis, and therapeutic resistance. Given their paramount importance, targeting CAFs emerges a robust strategy evolving landscape immunotherapy. Nanomaterials, distinguished by unique features malleability, hold considerable promise biomedicine, especially precision-oriented domain therapy. Their aptitude for modulating immune responses, amplifying drug efficacy through precise delivery, discerningly focusing on cells TME situates nanomaterials formidable tools to transcend boundaries set conventional treatments. This scrutinizes convoluted interplay among CAFs, cells, TME. It further showcases widely utilized management. We underscore potential nanoscale delivery systems directed at underscoring transformative power revolutionizing therapies, enhancing precision, culminating improved patient outcomes.

Despite remarkable progress in medical sciences, modern man is still fighting the battle against cancer. In 2022, only USA, 640 000 deaths and 2 370 patients were reported because of Chemotherapy most widely used for cancer treatments. However, chemotherapeutics have severe physicochemical side effects. Therefore, we prepared poly(amididoamine) dendrimeric carrageenan (CG), sodium alginate (SA) poly(vinyl alcohol) (PVA) hydrogels by using solution casting methodology. The constituents cross-linked mutable quantity 3-aminopropyl(diethoxy)methyl silane (APDMS). Hydrogels characterized Fourier transform infrared spectroscopy, thermal gravimetric analysis, scanning electron microscope atomic force microscopy. exhibited higher swelling volumes 5–7 pH range. vitro biodegradation ribonuclease-A cytocompatibility analysis DF-1 fibroblasts established their biodegradable non-toxic nature, which enables them as a suitable carrier chemotherapeutic compounds. Hence, methotrexate (MTX) model drug was loaded on CAP-8 hydrogel its release detected UV–visible spectrophotometer phosphate-buffered saline (PBS) solution. 13.5 h, 81.25% 77.23% MTX released at 7.4 (blood pH) 5.3 (tumour PBS, respectively. super case II mechanism best fitted to zero-order Korsmeyer–Peppas model. synthesized APDMS CG/SA/PVA could be an efficient platform effective delivery
