Previous studies have demonstrated an association of the NC_000012.12:g.53962605A > G, (rs2366152) single-nucleotide variant (SNV) situated in long noncoding homeobox transcript antisense intergenic RNA (HOTAIR) gene with HPV16-related cervical cancer pathogenesis. However, little is known about role rs2366152 progression and how oral birth control pills use, parity, menopausal status, cigarette smoking influence carcinogenesis. HRM analysis was used to determine SNV prevalence patients squamous cell carcinoma (SCC) (n = 470) group 499) a Polish Caucasian population. Logistic regression analyses were adjusted for age, using pills, smoking. Our genetic revealed that G/A vs. A/A (p 0.031, p 0.002) + G/G 0.035, 0.003) genotypes significantly related grade differentiation G3 tumor stage III, respectively. Moreover, risk increased among who smoked cigarettes pills. We conclude may promote invasion rapid growth SCC. can also be factor cancerogenesis.

The complex signaling network within the breast tumor microenvironment is crucial for its growth, metastasis, angiogenesis, therapy escape, stem cell maintenance, and immunomodulation. An array of secretory factors their receptors activate downstream cascades regulating cancer progression metastasis. Among various pathways, EGFR, ER, Notch, Hedgehog pathways have recently been identified as in terms proliferation, survival, differentiation, maintenance CSCs, failure. These mediate such MAPK, including MEK/ERK that promote common pro-oncogenic signaling, whereas dysregulation PI3K/Akt, Wnt/β-catenin, JAK/STAT activates key oncogenic events drug resistance, CSC enrichment, metabolic reprogramming. Additionally, these orchestrate an intricate interplay between stromal cells, immune cells. Metabolic reprogramming adaptations contribute to aggressive are unresponsive therapy. Glioblastoma multiforme (GBM) is an aggressive brain tumor. This meta-analysis investigates the association between HOTAIR expression levels and GBM. We searched literature for studies on in GBM patients. A of nine assessed standardized mean difference (SMD) 95% confidence intervals (CIs) using a random-effects model. Subgroup analyses were performed based sample source, country, study design. Additionally, we conducted meta-regression publication bias analyses. The found significant positive elevated (SMD = 8.3, CI 5.8-10.8, p 0.00). Considerable heterogeneity was observed (Q-value: 1174.2, df 9, I2 99.2%, indicated associations tissue samples from USA China. Meta-regression revealed that design country contributed to heterogeneity, with no detected. analysis confirms link GBM, highlighting as potential therapeutic target biomarker. Further research necessary clarify biological mechanisms involved this association.

Metabolic reprogramming plays a crucial role in cancer biology and the mechanisms underlying its regulation represent promising study area. In this regard, discovery of non-coding RNAs opened new regulatory landscape, which is early stages investigation. Using differential expression model HOTAIR, we evaluated level metabolic enzymes, as well metabolites produced by glycolysis glutaminolysis. Our results demonstrated effect HOTAIR on glutaminolysis enzymes colorectal cells. Specifically, through overexpression inhibition determined influence PFKFB4, PGK1, LDHA, SLC1A5, GLUD1, GOT1, had direct impact lactate glutamate production. Among effective herbal compounds cancer treatment, curcumin is known be one most effective. Curcumin has potential modulate critical pathways involved cell proliferation, apoptosis, cycle arrest, oxidative stress, epigenetic processes. Objectives: In this research, we investigated effects nanoparticles (curcumin-NPs) on expression DNMT3A, SUZ12, HOTAIR genes, MecP2 as important factors line. Methods: NPs were synthesized using ultrasonic methods, their toxic 1321N1 line measured 48 hours after MTT assay. RT-qPCR Western blot used measure levels different factors, including protein. Results: The results showed 50% inhibitory effect at IC50 concentration (1.7μM) curcumin-NPs prepared by method can reduce invasion, growth, migration Also, decrease was found HOTAIR, protein curcumin-NPs, which statistically (mean ± SD, n = 2, P < 0.05). Abstract Background Pancreatic adenocarcinoma, recognized for its aggressive behavior and frequent late-stage diagnosis, imposes significant challenges in early detection prognosis. This study aimed to evaluate the diagnostic prognostic potential by measuring expression levels of long non-coding RNA HOTAIR glycolytic enzyme hexokinase 2 (HK2) mRNA both tumorous adjacent non-tumorous pancreatic tissue samples ( n = 25 each) using RT-qPCR. Results Both lncRNA HK2 significantly increased tissues compared P 0.001). However, their stage T2 T3 showed no statistically difference 0.01). positively correlated with each other 0.001; r 0.95); however, associations were found serum tumor markers CA19-9 CEA 0.01; 005; p 0.1, 0.2). ROC analysis demonstrated abilities discriminate between (AUC 0.92 0.84, respectively) 96% 88% sensitivity, 72% 40% specificity, respectively, at optimal cut-off values 1.12 0.84 relative units. Patients elevated had shorter median survival (8 7 months, respectively), increasing risk adverse outcomes or recurrence 4–4.8 times (HR 4.08, 0.07; HR 4.8, 0.01), thus emphasizing cancer. Conclusion exhibit tumors. Elevated correlate strongly outcomes, underscoring value.
