Reports indicate that depression is a common mental health issue following traumatic brain injury (TBI). Our prior research suggests Nucleotide-binding oligomerization domain-like receptor protein 3 (NLRP3)-related neuroinflammation, modulated by glial cells such as astrocytes, likely to play crucial role in the progression of anxiety and cognitive dysfunction. However, there limited understanding potential astrocytic NLRP3 treating under mild TBI condition. This study aimed determine whether knockout (KO) could mitigate depressive-like behaviors explore variations between genders post-mild TBI. Mild was induced mice using Feeney's weight-drop method. This study explores a novel alternative exosome therapy, aimed at stimulating estrogen production in ovarian tissues, thus offering potential non-hormonal treatment for perimenopausal symptoms. Employing ex vivo methodologies, cortex specimens from women were treated exosomes derived human umbilical cord mesenchymal stem cells and cultured under specific conditions (patent number: PCT/US2022/073467). The produced cyclic guanosine monophosphate (cGMP) conditions, ensuring high safety standards. Estrogen levels quantified using enzyme-linked immunosorbent assay (ELISA), gene expression changes follicle-stimulating (FSH) receptors assessed via quantitative polymerase chain reaction (PCR).
