Abstract Background Growth differentiation factor 15 (GDF15) is a crucial biomarker in various physiological and pathological processes. While elevated GDF15 levels are linked to increased mortality risk, the role of DNA methylation (DNAm)-predicted predicting has not been extensively studied. The purpose study investigate association between DNAm-predicted all-cause cardiovascular disease (CVD) nationally representative cohort. The lack of accurate, cost-effective, and clinically relevant biomarkers remains a major barrier to incorporating omic data into clinical practice. Previous studies have shown that DNA methylation algorithms utility as surrogate measures for selected proteins metabolites. We expand upon this work by creating DNAm surrogates, termed epigenetic biomarker proxies (EBPs), across laboratories, the metabolome, proteome. After screening >2,500 biomarkers, we trained tested 1,694 EBP models assessed their incident relationship with 12 chronic diseases mortality, followed up 15 years. observe broad relevance: 1) there are 1,292 4,863 FDR significant prevalent associations, respectively; 2) most these associations replicated when looking at lab-based counterpart, > 62% shared higher odds hazard ratios disease outcomes than respective observed measurements; 3) EBPs current biochemistries detect deviations from normal high sensitivity specificity. Longitudinal also demonstrate changes corresponding in counterparts. Using two cohorts 30,000 individuals, found validate healthy sick populations. While further study is needed, findings highlight potential implementing simple, low-cost, high-yield framework benefits medicine.
