Alzheimer’s disease (AD) is a progressive neurodegenerative disorder characterized by cognitive impairment with amyloid-β (Aβ) accumulation, tau hyperphosphorylation, and neuroinflammation. Among these pathological features, microglial activation hallmark of Chaga (Inonotus obliquus) extract has been traditionally used for its diverse pharmacological properties, including anti-inflammatory neuroprotective effects. This study aimed to evaluate the therapeutic potential INO10, an inotodiol-rich chaga extract, in murine BV2 cells 3xTg-AD mouse model. In cells, INO10 significantly reduced LPS-induced expression pro-inflammatory cytokines (IL-1β, IL-6, TNF-α), indicating potent Oral administration improved spatial memory mice, as evidenced increased spontaneous alternation Y-maze test. Furthermore, treatment attenuated neuroinflammation, indicated downregulated cytokines. addition, immunohistochemical analysis confirmed that exhibited favorable bioavailability, supporting agent. Histological further revealed reduction Ab accumulation phosphorylation hippocampus, accompanied marked decrease neuroinflammatory markers. These findings suggest effectively mitigates AD-related pathology reducing Aβ deposition, ultimately leading enhancement. Given multi-target may serve promising natural compound AD treatment. Further investigations are warranted elucidate precise mechanisms clinical applicability.
